玉米ZmWRKY45基因克隆及鹽堿逆境脅迫下的表達分析

【摘要】:通過水稻中OsWRKY45基因在NCBI中進行同源序列比對,電子克隆ZmWRKY45基因并與其他物種中的同源基因進行進化樹分析。利用該基因的CDS區(qū)設(shè)計特異引物,在3葉期對吉單441進行不同濃度NaHCO_3處理,對玉米雜交種吉單441及其雙親自交系進行驗證。結(jié)果表明,在吉單441及其父本中含有ZmWRKY45,在鹽堿脅迫下吉單441中該基因隨著鹽堿濃度的升高而表達量升高,初步判斷該基因可能在吉單441的耐鹽堿過程中起到正向調(diào)控作用。
[Abstract]:OsWRKY45 gene in rice was sequenced in NCBI, then ZmWRKY45 gene was cloned and analyzed with the phylogenetic tree of other species. Specific primers were designed using the CDS region of this gene. Jidan 441 was treated with different concentrations of NaHCO_3 at 3 leaf stage, and the maize hybrid Jidan 441 and its parents inbred lines were tested. There was ZmWRKY45in Jidan 441 and its male parent. Under salt and alkali stress, the expression of Jidan 441 increased with the increase of saline-alkali concentration. It was preliminarily concluded that the gene might play a positive role in the saline-alkali tolerance of Jidan 441.
