【摘要】：三重基序蛋白(the tripartite motif-containing protein,TRIM)家族是一類結(jié)構(gòu)保守、進(jìn)化快速的E3泛素蛋白連接酶,參與諸多重要的生命過(guò)程。TRIM36是TRIM家族的一員,trim36編碼E3泛素蛋白連接酶。本研究在已完成的半滑舌鰨(Cynoglossus semilaevis)全基因組測(cè)序及轉(zhuǎn)錄組測(cè)序的基礎(chǔ)上,利用RACE技術(shù)克隆得到半滑舌鰨trim36基因(Gen Bank登錄號(hào):KU244470),序列全長(zhǎng)為2 899 bp,其中ORF為2 214 bp,編碼737個(gè)氨基酸,5'非翻譯區(qū)(untranslated regions,UTR)為289 bp,3'UTR為396 bp。生物信息學(xué)分析顯示,半滑舌鰨TRIM36蛋白包含1個(gè)鋅指結(jié)構(gòu),2個(gè)B-BOX類型鋅指蛋白結(jié)構(gòu)域(B-Box-type zinc finger,BBOX),1個(gè)卷曲螺旋結(jié)構(gòu),1個(gè)羧基末端的纖連蛋白Ⅲ(fibronectin typeⅢ,FN3)結(jié)構(gòu)域和1個(gè)雙特異性蛋白激酶spla和蘭尼堿受體(spla kinase and ryanodine receptor,SPRY)結(jié)構(gòu)域。此外,在不同脊椎動(dòng)物中,trim36基因與其鄰近的基因具有保守的共線性關(guān)系。半定量RT-PCR及q RT-PCR結(jié)果顯示,在半滑舌鰨不同組織中,trim36基因表達(dá)存在差異。trim36在卵巢中表達(dá)量最高,其次為雌、雄魚的腦中;不同發(fā)育時(shí)期中,trim36基因在120日齡半滑舌鰨性腺中開始表達(dá),且卵巢表達(dá)量顯著高于精巢(P0.05)。原位雜交結(jié)果顯示,trim36基因雜交信號(hào)主要出現(xiàn)在Ⅰ期和Ⅱ期卵母細(xì)胞。trim36時(shí)空表達(dá)特征表明,其在卵巢腔的形成及Ⅰ期、Ⅱ期卵母細(xì)胞的形成中起重要作用。結(jié)果表明,trim36在性別發(fā)生方面有一定的作用,這為進(jìn)一步研究魚類trim36基因功能及TRIM家族基因功能提供線索和參考。
[Abstract]:The tripartite motif-containing protein trimer family is a conserved and rapidly evolving E3 ubiquitin protein ligase. TRIM36 is a member of the TRIM family that encodes E3 ubiquitin protein ligase. This study is based on the complete genome sequencing and transcriptome sequencing of Cynoglossus semilaevis (Cynoglossus semilaevis). By using RACE technique, the accession number of Gen Bank of Cynoglossus semilaevis trim36 is 2 899 BP, in which ORF is 2 214 BP, encoding 737 amino acids 5'untranslated regions (UTRR) and 289 BP / 3UTR is 396 BP.
