ACAT1基因干擾對人結(jié)腸癌細胞增殖及侵襲轉(zhuǎn)移的影響

【摘要】:目的:利用小干擾RNA(si RNA)技術(shù)干擾酰基輔酶A-膽固醇;D(zhuǎn)移酶1(ACAT1)基因在人結(jié)腸癌HT29細胞中的表達,觀察ACAT1基因表達下降對于人結(jié)腸癌HT29細胞增殖和侵襲遷移能力的影響,了解ACAT1在結(jié)腸癌發(fā)生發(fā)展中的作用。方法:通過脂質(zhì)體Hiper Fect將ACAT1 si RNA轉(zhuǎn)染至人結(jié)腸癌HT29細胞中干擾ACAT1基因的表達,利用Real time PCR檢測轉(zhuǎn)染前后ACAT1基因表達的變化,利用ACAT1基因干擾前后的HT29細胞,采用CFSE染色法檢測HT29細胞增殖能力的變化;Transwell小室法檢測HT29細胞侵襲和遷移能力的變化。結(jié)果:ACAT1 si RNA轉(zhuǎn)染HT29細胞后ACAT1 m RNA的表達明顯下降,ACAT1 si RNA轉(zhuǎn)染后的HT29細胞與轉(zhuǎn)染前比較,增殖及侵襲轉(zhuǎn)移的能力均受到明顯抑制,差異有統(tǒng)計學(xué)意義(P0.05)。結(jié)論:ACAT1基因干擾使HT29細胞的增殖減慢,侵襲遷移能力降低,為結(jié)腸癌治療提供新的靶點。
[Abstract]:Aim: to investigate the effect of the decrease of ACAT1 gene expression on the proliferation, invasion and migration of human colon cancer HT29 cells by interfering with the expression of acyl coenzyme A- cholesterol acyltransferase 1 ActaT1 gene in human colon cancer HT29 cells by using small interfering RNA(si RNAs. Methods: ACAT1 si RNA was transfected into human colon cancer HT29 cells by liposome Hiper Fect to interfere with the expression of ACAT1 gene. Real time PCR was used to detect the changes of ACAT1 gene expression before and after transfection. Using ACAT1 gene to interfere with HT29 cells, The changes of proliferation ability of HT29 cells were detected by CFSE staining and the changes of invasion and migration of HT29 cells were detected by Transwell chamber assay. Results the expression of ACAT1 m RNA in HT29 cells transfected with HT29 cells was significantly decreased after transfection of HT29 cells by RNA. The expression of ACAT1 m RNA in HT29 cells after transfection of HT29 cells was significantly lower than that in HT29 cells after transfection with ACAT1si RNA. The ability of proliferation and invasion and metastasis were significantly inhibited, and the difference was statistically significant (P 0.05). Conclusion the interference of the HT29 gene can slow down the proliferation and decrease the ability of invasion and migration of HT29 cells, which provides a new target for colon cancer therapy.
