敦煌石室大寶膠囊對(duì)衰老大鼠血清白細(xì)胞介素-4、-10含量和海馬區(qū)熱休克蛋白86基因蛋白表達(dá)的影響

【摘要】:目的觀察以健脾補(bǔ)腎-滌痰祛瘀法為指導(dǎo)的敦煌石室大寶膠囊(DHDB)對(duì)衰老模型大鼠血清白細(xì)胞介素(IL)-4、IL-10含量和海馬區(qū)熱休克蛋白(HSP)86基因和蛋白表達(dá)的影響。方法受試動(dòng)物按隨機(jī)數(shù)字表法分為空白組(n=10)和模型組(n=50)。采用D-半乳糖(0.125 g/kg)皮下注射建立衰老模型并篩選符合衰老模型的大鼠,按隨機(jī)數(shù)字表法分為模型組、DHDB高、中、低劑量組(0.8、0.4、0.2 g/kg)和陽(yáng)性對(duì)照組(0.4 g/kg吡拉西坦),經(jīng)藥物干預(yù)后分別測(cè)定各組大鼠血清IL-4、IL-10含量和海馬區(qū)HSP86基因和蛋白表達(dá)水平。結(jié)果與空白組比較,模型組大鼠學(xué)習(xí)能力下降,血清IL-4含量顯著降低、IL-10含量顯著增高,海馬區(qū)HSP86基因和蛋白表達(dá)水平顯著降低(均P0.05);DHDB各劑量組大鼠學(xué)習(xí)能力提高,血清IL-4含量顯著增高而IL-10含量顯著降低,海馬區(qū)HSP86基因和蛋白表達(dá)水平顯著增高,且以高劑量DHDB作用為優(yōu)(P0.05)。結(jié)論 DHDB具有提高衰老大鼠學(xué)習(xí)記憶能力,調(diào)節(jié)血清IL-4、IL-10含量正常分泌及促進(jìn)海馬區(qū)HSP86基因和蛋白表達(dá)的作用。
[Abstract]:Objective to observe the effect of DHDBs of Dunhuang Shizhuangdabao capsule (DHDBs) on serum interleukin IL-4 in aging model rats guided by the method of invigorating spleen and kidney and removing phlegm from phlegm and removing blood stasis. Effects of IL-10 content and heat shock protein (HSPN) 86 gene and protein expression in hippocampal region. Methods the tested animals were randomly divided into two groups: control group (n = 10) and model group (n = 50). The aging model was established by subcutaneous injection of D-galactose 0.125 g / kg and the aging model was selected. According to the method of random number table, the model group was divided into three groups: model group with high, medium and low doses of 0.8g / kg 0.40.2g / kg) and positive control group with 0.4 g / kg Piracetam). Serum IL-4 IL-10 level and HSP86 gene and protein expression level in hippocampus were measured after drug intervention. Results compared with the blank group, the learning ability of the model group decreased. The level of serum IL-4 decreased significantly, the level of IL-10 increased significantly, and the expression of HSP86 gene and protein in hippocampus decreased significantly (all P 0.05). The learning ability of rats in each dose group of DHDB was improved, the content of serum IL-4 was significantly increased, the content of IL-10 was significantly decreased, and the expression of HSP86 gene and protein was significantly increased in hippocampus. Conclusion DHDB can improve learning and memory ability and regulate serum IL-4 in aging rats. Normal secretion of IL-10 and promotion of HSP86 gene and protein expression in hippocampus.
