實(shí)時(shí)熒光定量RT-PCR檢測(cè)賴草LRC1基因表達(dá)研究

【摘要】:通過亞克隆技術(shù)獲得賴草LRC1基因片段重組質(zhì)粒,以其梯度稀釋樣品為標(biāo)準(zhǔn),建立了實(shí)時(shí)熒光定量PCR技術(shù)檢測(cè)LRC1基因絕對(duì)定量表達(dá)方法。進(jìn)一步運(yùn)用該技術(shù)和相對(duì)定量法對(duì)賴草初花期LRC1基因表達(dá)特性進(jìn)行分析發(fā)現(xiàn),該基因在根莖尖、根莖節(jié)間、根莖節(jié)、地上莖中的拷貝數(shù)分別為477.79、162.65、1368.00、53.86個(gè)拷貝/mg(組織鮮重);相對(duì)定量法檢測(cè)結(jié)果顯示,LRC1基因在賴草根莖節(jié)間中的表達(dá)量約為其在根莖節(jié)中表達(dá)量的10%,穗、葉片和莖中表達(dá)量則為其在根莖節(jié)中的1%、2%和1%。表明該基因在賴草根莖中的表達(dá)量顯著高于地上組織,特別是其在根莖芽產(chǎn)生的根莖節(jié)內(nèi)的表達(dá)量顯著高于其他組織。
[Abstract]:The recombinant plasmid of LRC1 gene fragment of Leymus chinensis was obtained by subcloning technique, and its gradient dilution was used as the standard. A real-time fluorescence quantitative PCR method was established to detect the absolute quantitative expression of LRC1 gene, and the characteristics of LRC1 gene expression in the early flowering stage of Rhizoma Raigrass were analyzed by using this technique and the relative quantitative method. The number of copies of the gene in rhizome tip, rhizome Internode, rhizome node and upper stem were 477.79 ~ 162.65.1368.00,53.86 copies / mg (fresh tissue weight), respectively. The results of relative quantitative analysis showed that the expression of LRC1 gene in rhizome Internode of Leymus chinensis was about 10% of that in rhizome node, and that in ear, leaf and stem was 1% of that in rhizome node. 2% and 1.The results showed that the expression of the gene in the rhizome of Leymus chinensis was significantly higher than that in the aboveground tissue, especially in the rhizome node produced by the rhizome bud.
