RNPC1與p53基因在肺腺癌組織中的表達及其臨床意義

【摘要】:目的:觀察肺腺癌組織中RNPC1、p53基因表達及其與肺腺癌臨床特征之間的關系,探討其在肺腺癌的發(fā)生發(fā)展中的意義。方法:采用RT-PCR和Western blot法檢測50例肺腺癌組織及對應癌旁組織中RNPC1、p53 mRNA及蛋白水平的相對表達量,分析其與肺腺癌臨床病理特征之間的關系,及其兩者之間的關系。結果:肺腺癌RNPC1 mRNA及蛋白相對表達量(0.357±0.170、0.294±0.149)均高于癌旁組織(0.271±0.128、0.206±0.099),肺腺癌組織p53 mRNA及蛋白(0.457±0.208、0.671±0.200)相對表達量均高于癌旁組織(0.308±0.167、0.332±0.071),差異有統(tǒng)計學意義(P0.01)。RNPC1表達與肺腺癌患者TNM分期、浸潤深度有關(P0.05)。p53表達與患者TNM分期有關(P0.05)。實驗組RNPC1與p53蛋白表達相關(r=-0.626,P0.01)。結論:肺腺癌組織RNPC1、p53 mRNA及蛋白表達高于癌旁組織,兩者均與患者部分病理參數(shù)關系密切。隨著RNPC1基因表達的增加p53蛋白隨之減少,RNPC1可能通過抑制p53的翻譯從而促進肺癌的發(fā)生發(fā)展。RNPC1可能成為肺腺癌分子靶向治療的靶點。
[Abstract]:Objective: to investigate the relationship between the expression of RNPC1 and p53 gene and the clinical features of lung adenocarcinoma. Methods: RT-PCR and Western blot were used to detect RNPC1 in 50 cases of lung adenocarcinoma and its adjacent tissues. The relative expression of p53 mRNA and protein, and the relationship between p53 mRNA and clinicopathological features of lung adenocarcinoma were analyzed. Results: the relative expression of RNPC1 mRNA and protein in lung adenocarcinoma was 0.357 鹵0.170. 0.294 鹵0.149) was higher than that of paracancerous tissue (0.271 鹵0.128 鹵0.206 鹵0.099). The relative expressions of p53 mRNA and protein in lung adenocarcinoma were higher than those in paracancerous tissues (0.457 鹵0.208 鹵0.671 鹵0.200). The difference was statistically significant between the expression of P0.01P. RNPC1 and the TNM stage of lung adenocarcinoma. The depth of invasion was related to the expression of P0.05, p53 and the TNM stage of the patients. The expression of RNPC1 and p53 protein was correlated with r-0.626 in the experimental group. Conclusion: the expression of p53 mRNA and protein in lung adenocarcinoma tissue is higher than that in paracancerous tissue. The expression of p53 protein decreased with the increase of RNPC1 gene expression. RNPC1 may promote the development of lung cancer by inhibiting the translation of p53. RNPC1 may be the target of molecular targeted therapy for lung adenocarcinoma.
