水稻HSP90基因家族功能研究

【摘要】:隨著近年來極端天氣的出現(xiàn)愈加頻繁,高溫、低溫、干旱及土壤鹽漬化等逆境脅迫對作物的生長、發(fā)育、產(chǎn)量的影響也越發(fā)凸顯。研究作物耐逆機制,培育耐逆品種成為當(dāng)前亟待解決的問題。HSP90家族蛋白是一類豐富的分子伴侶蛋白,并廣泛存在于真核生物中的保守蛋白。HSP90家族蛋白的主要功能主要是協(xié)助其它蛋白的正確折疊,并在逆境應(yīng)答網(wǎng)絡(luò)中發(fā)揮重要作用。本實驗中利用生物信息學(xué)的工具,發(fā)現(xiàn)并分析了日本晴中HSP90家族的9個OsHSP90蛋白(OsHSP90-1至OsHSP90-9).通過序列比對,我們在水稻HSP90家族蛋白中確定了三個基序。通過進化樹分析,將水稻HSP90家族蛋白分為三簇(Ⅰ,Ⅱ,Ⅲ)。本實驗在水稻10個組織中對這9個OsHSP90蛋白基因進行了表達量分析。結(jié)果表明在10個部位的組織中OsHSP90-4、OsHSP90-6和OsHSP90-7有較高的表達水平,而OsHSP90-5和OsHSP90-8的表達水平非常低。OsHSP90-1只特異性的在開花14天后的胚中表達。據(jù)文獻報道,在熱激和其它逆境脅迫下,熱激蛋白的表達會受到正向調(diào)控。但在本研究中,不同逆境脅迫條件下,水稻OsHSP90家族蛋白的表達模式卻是各式各樣的。在干旱、高鹽、低溫、熱激脅迫下OsHSP90-2和OsHSP90-4的表達受到了正向調(diào)控,在高鹽、干旱的脅迫下OsHSP90-3和OsHSP90-5的表達受到了負向調(diào)控。在干旱脅迫下OsHSP90-7和OsHSP90-9受到負向調(diào)控。與對照相比,在多種逆境脅迫下OsHSP90-6的表達量基本保持不變。我們分別用OsHSP90-2和OsHSP90-4轉(zhuǎn)化大腸桿菌,均能提高轉(zhuǎn)化菌株的細胞活性并提高對高溫、高鹽、干旱逆境環(huán)境的耐受力。我們分別用OsHsP90-2和OSHsP90-4轉(zhuǎn)化水稻,但只有OsHSP90-4的水稻轉(zhuǎn)化子對高溫的耐受力有明顯提高并增強水稻轉(zhuǎn)化子對ABA的敏感性。這些結(jié)果為進一步研究水稻OsHSP90基因家族提供了新的視角。
