不同溫度對牛精子相關(guān)抗原11D基因的影響

【摘要】:為探討高溫對精子膜蛋白牛精子相關(guān)抗原11D(sperm associated antigen 11,SPAG11D)表達(dá)水平的影響,試驗采用了實時熒光定量PCR和Western boltting方法檢測了牛睪丸原代細(xì)胞和娟姍牛、檳榔江牛、西門塔爾牛的正常精子及低活力精子中SPAG11D的表達(dá)。結(jié)果顯示,牛睪丸原代細(xì)胞在不同溫度(32、34、36、38和40℃)下培養(yǎng)24h,SPAG11D的mRNA及蛋白在36℃時表達(dá)水平最高,且高溫時表達(dá)水平比低溫時要高;培養(yǎng)48h時,SPAG11D的mRNA表達(dá)基本和24h相一致,但蛋白表達(dá)水平在高溫時明顯下降。此外,檳榔江牛和娟姍牛低活力精子中SPAG11D的蛋白表達(dá)量比正常精子明顯降低,而在西門塔爾牛的精液中變化不明顯。
[Abstract]:To investigate the effect of high temperature on the expression of spermatozoa associated antigen (11DX) in bovine spermatozoa associated protein (SPAG11D). Real time fluorescence quantitative PCR and Western boltting were used to detect the primary cells of bovine testis and betel nut cattle. Expression of SPAG11D in normal and low motility spermatozoa of Simmental cattle. The results showed that primary bovine testicular cells were cultured at different temperatures for 24 hours.
