【摘要】：【目的】在100名智力障礙患者中篩查基因POGZ的突變,探索POGZ的突變與智力障礙的關(guān)系�！痉椒ā刻崛』颊咄庵苎狣NA,對(duì)POGZ的外顯子、外顯子-內(nèi)含子接頭區(qū)以及5’UTR區(qū)、3’UTR區(qū)進(jìn)行PCR-測(cè)序分析。通過(guò)與數(shù)據(jù)庫(kù)中數(shù)據(jù)進(jìn)行比對(duì),查找突變位點(diǎn)�！窘Y(jié)果】在100名智力障礙患者中檢測(cè)出一例未報(bào)道過(guò)的新錯(cuò)義突變c.2498 GA,p.H833R。該突變破壞了POGZ蛋白上與HP1結(jié)合起關(guān)鍵作用的C2H2鋅指結(jié)構(gòu),可能使POGZ無(wú)法與HP1蛋白結(jié)合進(jìn)而影響其調(diào)控細(xì)胞周期的功能�！窘Y(jié)論】POGZ基因的突變是引起先天性智力障礙的原因之一。對(duì)不明原因智力障礙的患者做POGZ的篩查有助于進(jìn)一步明確病因,對(duì)有智力障礙家族史的孕婦做產(chǎn)前檢查能防止患兒的出生。
[Abstract]:[objective] to screen the mutation of POGZ gene in 100 patients with mental retardation and to explore the relationship between the mutation of POGZ and mental retardation. [methods] DNA was extracted from peripheral blood of the patients. The exon, exon-intron junction region and 5UR region of POGZ were analyzed by PCR-sequencing. The data were compared with the data in the database. [results] one unreported new missense mutation c.2498 GA was detected in 100 patients with mental retardation. P. H833R. this mutation destroyed the finger structure of C _ 2H _ 2 zinc finger which plays a key role in the binding of POGZ protein to HP1. It may be that POGZ can't bind to HP1 protein and affect its function of regulating cell cycle. [conclusion] the mutation of POGZ gene is one of the causes of congenital mental retardation. Screening for POGZ in 10% of patients is helpful in further understanding the etiology. Prenatal examination of pregnant women with a family history of mental retardation can prevent the birth of their children.
