【摘要】：目的:DNA損傷修復(fù)基因在癌癥的發(fā)生發(fā)展過程中起著至關(guān)重要的作用。本課題對我國人群7個DNA損傷修復(fù)基因中32個單核苷酸多態(tài)性位點(SNP)在膀胱癌臨床發(fā)生發(fā)展過程中所起的作用及與預(yù)后之間的關(guān)系進行研究。方法:應(yīng)用TaqMan OpenArray基因芯片技術(shù)對我國227名膀胱癌患者進行了高通量基因分型檢測,分析其與膀胱癌預(yù)后相關(guān)因素的關(guān)系。結(jié)果:在有吸煙習慣的膀胱癌患者中APEX1基因rs1130409位點SNP與膀胱癌的易感性相關(guān)(P0.001);XRCC1基因的rs3213356位點、APEX1基因rs3136817位點以及MUTYH基因的rs3219493位點不同SNP即變異型與野生純合子與膀胱癌的臨床病理特征具有相關(guān)性(P0.001;P0.001;P0.001);APEX1基因rs3136817位點和XRCC2基因rs3218454位點SNP可預(yù)測患者復(fù)發(fā)風險(P0.001;P0.001),常見于變異基因型當中。PARP1基因rs1805415位點的變異基因型相比野生型,生存時間相對較長(P=0.02;P=0.04)。結(jié)論:DNA損傷修復(fù)基因多個單核苷酸多態(tài)性位點膀胱癌患者的臨床病理特征和預(yù)后具有顯著相關(guān)性。
[Abstract]:Objective to study the role of DNA damage repair gene in the development of cancer. This study has been applied to 32 single nucleotide polymorphisms (SNPs) of 7 DNA damage repair genes in Chinese population. Methods: to study the role of TaqMan in the clinical development of bladder cancer and its relationship with prognosis. High-throughput genotyping of 227 bladder cancer patients in China was detected by OpenArray gene chip technique. Results: in patients with bladder cancer with smoking habit, the SNP of rs1130409 locus of APEX1 gene was associated with the susceptibility of bladder cancer. P0.001; The rs3213356 locus of XRCC1 gene. The clinicopathological characteristics of APEX1 gene rs3136817 locus and MUTYH gene rs3219493 locus are correlated with the clinicopathological characteristics of bladder cancer. (. P0.001; P0.001; P0.001; SNP at rs3136817 site of APEX1 gene and rs3218454 locus of XRCC2 gene could predict the recurrence risk of patients (P0.001). P0.001A, the survival time of the variant genotypes in the rs1805415 locus of the parP1 gene was longer than that of the wild genotype. The survival time of P0. 02 was longer than that of the wild genotype at the rs1805415 locus of PARP1 gene. Conclusion there is a significant correlation between clinicopathological features and prognosis of bladder cancer patients with multiple single nucleotide polymorphisms of DNA damage repair gene.
