【摘要】:目的:通過篩選差異基因,獲得控制骨髓間充質(zhì)干細(xì)胞向神經(jīng)細(xì)胞分化及神經(jīng)發(fā)育的中心基因,為治療神經(jīng)系統(tǒng)疾病提供參考。方法:從基因表達(dá)綜合數(shù)據(jù)庫(Gene Expression Omnibus database)中獲得芯片數(shù)據(jù),利用生物信息學(xué)軟件篩選差異基因,并對差異基因進(jìn)行GO功能富集、蛋白互作網(wǎng)絡(luò)分析和中心基因分析。結(jié)論:通過分析,初步推測Nrcam、Sema3a、Mapk8、Dlg4、Slit1、Creb1、Ntrk2、Cntn2和Pax6等中心基因在調(diào)控骨髓間充質(zhì)干細(xì)胞向神經(jīng)細(xì)胞的分化中發(fā)揮重要作用;Dcx、Nrcam、Sema3a、Cntn2、Slit1、Ephb1和Pax6等中心基因在神經(jīng)發(fā)育過程中發(fā)揮作用;Fgf2、Tgfβ1、Vegfa、Serpine1、Il6和Stat1等中心基因在抑制神經(jīng)分化過程中發(fā)揮作用。
[Abstract]:Objective: to obtain the central gene to control the differentiation and neural development of bone marrow mesenchymal stem cells (BMSCs) by screening differentially expressed genes. Methods: microarray data were obtained from Gene Expression Omnibus database. The differential genes were screened by bioinformatics software, and the differential genes were enriched with go function, protein interaction networks and central genes were analyzed. Conclusion: through the analysis, we preliminarily speculate Nrcam. Sema3a,Mapk8,Dlg4,Slit1,Creb1,Ntrk2. Central genes such as Cntn2 and Pax6 play an important role in regulating the differentiation of bone marrow mesenchymal stem cells into neural cells. Cntn2Slit1, Ephb1 and Pax6 genes play a role in neurodevelopment. Fgf2tgf 尾 1, Vegfa1, Serpine 1, Stat1 and other central genes play an important role in the inhibition of neural differentiation.
