【摘要】：目的探討人類白細(xì)胞抗原(human leukocyte antigen,HLA)DP基因多態(tài)性與中國人群特發(fā)性膜性腎病(Idiopathic Membranous Nephropathy,IMN)遺傳易感性的關(guān)系。方法選取185例特發(fā)性膜性腎病患者和405位健康對照作為研究對象,應(yīng)用Taq Man-MGB探針基因分型方法檢測HLA-DP基因多態(tài)性位點(diǎn)(rs3077和rs9277535)的基因型分布。結(jié)果采用多因素logistic回歸分析,rs3077位點(diǎn)結(jié)果顯示,突變基因型AA在特發(fā)性膜性腎病組中的頻率均顯著高于健康對照組(調(diào)整OR=1.92,95%CI=1.09-3.38);單倍型分析顯示,與GG單倍型相比,攜帶AA單倍型的個體可增加特發(fā)性膜性腎病的患病風(fēng)險;(調(diào)整OR=2.86,95%CI=1.58-5.58);此外,rs3077位點(diǎn)的突變基因型AA可增加特發(fā)性膜性腎病患者發(fā)生腎功能衰竭的風(fēng)險(調(diào)整OR=3.11,95%CI=1.53-6.32)。結(jié)論 HLA-DP基因多態(tài)性與特發(fā)性膜性腎病遺傳易感性存在關(guān)聯(lián),且與特發(fā)性膜性腎病患者的腎功能也具有相關(guān)性。
[Abstract]:Objective to investigate the human leukocyte antigen of human leukocyte antigen. HLA)DP gene polymorphism and idiopathic Membranous Nephropathy in Chinese patients with idiopathic membranous nephropathy. Methods 185 patients with idiopathic membranous nephropathy and 405 healthy controls were selected. Detection of polymorphic HLA-DP loci rs3077 and rs9277535 by Taq Man-MGB probe genotyping. Results multivariate logistic regression analysis was used. The results of rs3077 locus showed that the frequency of AA mutation genotype in idiopathic membranous nephropathy group was significantly higher than that in healthy control group (adjusted OR=1.92). 95. CII 1.09-3.38; Haplotype analysis showed that individuals with AA haplotype increased the risk of idiopathic membranous nephropathy compared with GG haplotype. (adjustment of ORA 2.86 / 95 / CI1.58-5.58); In addition, the mutation genotype AA at rs3077 increased the risk of renal failure in patients with idiopathic membranous nephropathy (adjusted for OR=3.11). Conclusion the polymorphism of HLA-DP gene is associated with the genetic susceptibility of idiopathic membranous nephropathy. It is also associated with renal function in patients with idiopathic membranous nephropathy.
