咖啡堿和茶多酚組合對(duì)小鼠脂肪酸合成酶基因和蛋白表達(dá)的影響

【摘要】:探討咖啡堿和茶多酚組合對(duì)小鼠肝臟脂肪酸合成酶(FAS)基因和蛋白表達(dá)的影響,闡明其減肥機(jī)制。50只昆明小鼠隨機(jī)被分成5組,分別為對(duì)照組(I)、0.03%咖啡堿+0.3%茶多酚(II)、0.03%咖啡堿+0.6%茶多酚(III)、0.06%咖啡堿+0.3%茶多酚(IV)、0.06%咖啡堿+0.6%茶多酚(V),喂養(yǎng)12周。分析肝臟FAS的m RNA和蛋白表達(dá)量?Х葔A與茶多酚組合的小鼠體重增加與對(duì)照組相比均有下降趨勢(shì),特別是V組的小鼠體重增加顯著減少,咖啡堿與茶多酚組合的小鼠腹腔內(nèi)脂肪(IPAT)質(zhì)量均明顯減少。IV、V組的血中TG、TC濃度顯著降低,II、III組的NEFA濃度明顯降低。與對(duì)照組相比,咖啡堿和茶多酚組合FAS m RNA表達(dá)量均顯著降低,IV和V組的FAS蛋白表達(dá)量顯著減少。以上結(jié)果表明咖啡堿與茶多酚組合通過調(diào)控小鼠FAS的m RNA及蛋白表達(dá)量,抑制脂肪合成,降低血脂濃度,引起體內(nèi)脂肪沉積減少,達(dá)到預(yù)防肥胖的效果。
[Abstract]:To investigate the effect of caffeine and tea polyphenols on the expression of FASgene and protein in mouse liver fatty acid synthase, 50 Kunming mice were randomly divided into 5 groups. The control group was 0.03% caffeine 0.3% tea polyphenols, 0.03% caffeine 0.6% tea polyphenols. 0.06% caffeine 0.3% tea polyphenols 0.06% caffeine 0.6% tea polyphenols V). After 12 weeks of feeding, the expression of m RNA and protein in liver FAS was analyzed. The weight gain of mice combined with caffeine and tea polyphenols showed a decreasing trend compared with the control group. Especially the weight gain of mice in group V was significantly decreased, and the weight of IP ATA of mice combined with caffeine and tea polyphenols was significantly decreased. The concentration of TGN TC in blood of group V was significantly lower than that of group V. Compared with the control group, the expression of FAS m RNA in caffeine and tea polyphenols was significantly decreased. The results showed that the combination of caffeine and tea polyphenols could inhibit fat synthesis and decrease the concentration of lipid by regulating the expression of m RNA and protein of FAS in mice. Causes the body fat deposition to reduce, achieves the prevention obesity effect.
