【摘要】：為了研究Wnt6基因在簡州大耳羊不同器官組織中的表達水平,試驗采用RT-PCR方法克隆山羊Wnt6基因序列,采用熒光定量PCR技術檢測該基因在各器官組織中的表達情況。結(jié)果表明:獲得山羊Wnt6基因序列1 120 bp(Gen Bank登錄號為KU950833),其中CDS為1 098 bp,5'UTR16 bp和3'UTR 6 bp,編碼365個氨基酸,存在1個信號肽剪切位點,跨膜序列為7~29位氨基酸。氨基酸同源性比較發(fā)現(xiàn),山羊Wnt6與綿羊和牛的同源性最高達99%;山羊Wnt6基因在心臟、肝臟、脾臟、肺臟、腎臟、脂肪、背最長肌、股二頭肌、臂三頭肌中存在廣泛表達,且在脂肪組織中的表達水平極顯著高于其他器官組織(P0.01)。
[Abstract]:In order to study the expression level of Wnt6 gene in different organs and tissues of large ear sheep in Jianzhou, RT-PCR method was used to clone goat Wnt6 gene sequence. Fluorescence quantitative PCR technique was used to detect the expression of the gene in various organs and tissues. The results showed that the goat Wnt6 gene sequence was 1 120 BP (. Gen Bank login number is KU950833). The CDS was 1 098 BP / 5 UTR16 BP and 3 UTR6 BP, encoding 365 amino acids, and there was a signal peptide splicing site. The transmembrane sequence was 7 ~ 29 amino acids. The homology of amino acid homology between goat Wnt6 and sheep and cattle was 999.The results showed that goat Wnt6 had the highest homology with sheep and cattle. Goat Wnt6 gene is widely expressed in heart, liver, spleen, lung, kidney, fat, longissimus dorsi muscle, biceps femoris muscle and triceps arm muscle. The expression level in adipose tissue was significantly higher than that in other organs.
