利用Pigm基因改良粳稻保持系的稻瘟病抗性研究

【摘要】:選育和應(yīng)用抗病品種是防治水稻稻瘟病經(jīng)濟有效的技術(shù)途徑。為了改良粳稻保持系浙04B的稻瘟病抗性,以含稻瘟病廣譜抗性基因Pigm的粳稻材料MP3為供體親本,浙04B為受體親本,通過雜交、回交并結(jié)合分子標記輔助選擇,將Pigm基因?qū)胝?4B中,利用分布于水稻12條染色體上的47個多態(tài)性SSR標記檢測近等基因系的遺傳背景回復(fù)率,獲得了23份背景回復(fù)率0.80的近等基因系。選用4個浙江省稻瘟病優(yōu)勢菌株Zj-5、Zj-8、13-37和13-41人工接種鑒定,23份近等基因系均表現(xiàn)為抗穗頸瘟。田間自然誘發(fā)鑒定結(jié)果表明,23份近等基因系葉瘟為0級,穗瘟均為1級,表現(xiàn)為抗穗瘟,表明近等基因系稻瘟病抗性明顯提高。對10項農(nóng)藝性狀的考察結(jié)果表明,導(dǎo)入Pigm基因的近等基因系劍葉變短,千粒重增加,生育期延遲,株高增加,有效穗數(shù)、每穗總粒數(shù)和產(chǎn)量總體減少。本研究結(jié)果為選育抗稻瘟病新品種提供了材料,同時為Pigm基因抗病育種應(yīng)用提供了參考。
[Abstract]:Breeding and application of disease-resistant varieties is an economical and effective technical way to control rice blast in order to improve rice blast resistance of japonica rice maintainer line Zhejiang 04B. Japonica rice (MP3) containing the broad spectrum resistance gene (Pigm) of rice blast was used as donor parent and Zhejiang 04B as recipient parent. The Pigm gene was introduced into Zhejiang 04B and 47 polymorphic SSR markers distributed on 12 rice chromosomes were used to detect the recovery rate of genetic background of near-isogenic lines. 23 near-isogenic lines with a response rate of 0.80 were obtained. Four dominant strains of rice blast in Zhejiang Province, Zj-5, Zj-8J-83, 13-37 and 13-41, were identified by artificial inoculation. All the 23 near-isogenic lines showed resistance to panicle blast. The results of field induced identification showed that leaf blast of 23 near-isogenic lines was grade 0 and panicle blast was grade 1, which showed resistance to panicle blast. The results showed that the resistance of near-isogenic lines to rice blast was significantly increased. The results of 10 agronomic traits showed that the leaf of the near-isogenic lines with Pigm gene was shorter, the 1000-grain weight was increased, the growth period was delayed, and the plant height was increased. The number of effective panicles, the total grains per panicle and the yield were decreased. The results provided a reference for breeding new varieties resistant to rice blast and for the application of Pigm gene in disease resistance breeding.
