人宮頸癌Hela細(xì)胞TPX2基因沉默對(duì)放射敏感性的影響

【摘要】:目的觀察TPX2基因沉默的人宮頸癌Hela細(xì)胞對(duì)X射線的敏感性。方法將宮頸癌Hela細(xì)胞分為T(mén)PX2-siRNA組、NC-siRNA組、Control組,TPX2-siRNA組轉(zhuǎn)染TPX2-siRNA,NC-siRNA組轉(zhuǎn)染NC-siRNA即空載體siRNA,Control組僅加入轉(zhuǎn)染試劑。采用實(shí)時(shí)熒光定量PCR、Western blot分別檢測(cè)各組細(xì)胞中的TPX2 mRNA與蛋白。采用CCK8法檢測(cè)不同劑量(0、2、4、6 Gy)X線照射后TPX2-siRNA組細(xì)胞的增殖抑制率。采用流式細(xì)胞術(shù)檢測(cè)4 Gy X線照射前后TPX2-siRNA組細(xì)胞的凋亡率。結(jié)果與NC-siRNA組、Control組相比,TPX2-siRNA組細(xì)胞TPX2 mRNA、蛋白相對(duì)表達(dá)量降低(P均0.05)。TPX2-siRNA組細(xì)胞接受0、2、4、6 Gy X線照射后,細(xì)胞增殖抑制率分別為4.71%±0.15%、5.26%±0.69%、6.64%±0.13%、15.19%±0.73%,兩兩組間比較P均0.05。TPX2-siRNA組細(xì)胞接受4 Gy X線照射前后細(xì)胞凋亡率分別為11.43%±0.03%、12.68%±0.03%,兩者比較P0.05。結(jié)論 TPX2基因沉默的人宮頸癌Hela細(xì)胞對(duì)X射線的敏感性增加。
[Abstract]:Objective to observe the radiosensitivity of human cervical cancer Hela cells silenced with TPX2 gene to X-ray. Methods Cervical cancer Hela cells were divided into TPX2-siRNA group and NC-siRNA group. Control group was transfected with TPX2-siRNA group and NC-siRNA group was transfected into NC-siRNA vector siRNA. In Control group, only transfection reagent was added and real-time quantitative PCR was used. Western blot was used to detect the TPX2 mRNA and protein in the cells of each group, and different doses of TPX2 mRNA and protein were detected by CCK8 method. 6 Gy). The proliferation inhibition rate of TPX2-siRNA group after X-ray irradiation was determined by flow cytometry. The apoptosis rate of TPX2-siRNA group before and after X-ray irradiation was compared with that of NC-siRNA group. Compared with the TPX2 mRNAs of the Control group, the relative protein expression of the Control group was lower than that of the TPX2-siRNA group. The cells in the TPX2-siRNA group received 0. 2After X ray irradiation, the inhibition rate of cell proliferation was 4.71% 鹵0.15 and 5.26% 鹵0.69 and 6.64% 鹵0.13%, respectively. 15.19% 鹵0.73%. The apoptosis rate of the cells in the TPX2-siRNA group was 11.43% 鹵0.03% before and after 4 Gy X-ray irradiation. 【正文快照】: TPX2是一種編碼基因定位于人12號(hào)染色體的微管相關(guān)蛋白,能激活細(xì)胞周期激酶Aurora A,并調(diào)節(jié)有絲分裂紡錘體的形成[1]。已有研究[2]發(fā)現(xiàn),TPX2過(guò)表達(dá)于多種腫瘤細(xì)胞內(nèi),且與染色體的不穩(wěn)定性密切相關(guān)。在女性惡性腫瘤中,宮頸癌的發(fā)病率逐年上升,目前放療已經(jīng)成為宮頸癌的主要治療
