針刺對抑郁癥大鼠海馬5-HT、NE、BDNF水平和神經(jīng)元細胞凋亡相關基因表達的影響

【摘要】:目的:探討針刺對神經(jīng)元細胞的保護作用及其可能分子機制。方法:側腦室注射鵝膏蕈氨酸建立神經(jīng)元損傷大鼠模型;造模成功后,給予針刺和藥物干預,觀察大鼠的行為學變化、海馬神經(jīng)元細胞形態(tài)學改變;檢測單胺類神經(jīng)遞質(5-HT、NE)含量的變化、腦源性神經(jīng)因子(BDNF)和凋亡蛋白Bax/Bcl-2的變化。結果:與空白組比較,模型組表現(xiàn)出運動量減少(P0.05)、食欲和對新環(huán)境的適應能力下降(P0.05)、神經(jīng)元細胞數(shù)目減少、核內空泡增多、連接松散、5-HT和NE含量顯著下降(P0.05)、Bax/Bcl-2比值升高(P0.05)。針刺和藥物干預后,大鼠運動量、食欲和對新環(huán)境的適應能力得到加強(P0.05),神經(jīng)元細胞數(shù)目增加,連接松散的形態(tài)得到改善,核內空泡數(shù)量減少,5-HT、NE和BDNF含量均升高(P0.05),Bax/Bcl-2比值降低(P0.05)。結論:針刺對神經(jīng)元的保護作用可能是通過提高5-HT、NE和BDNF的含量,減少神經(jīng)元細胞凋亡起作用的。
[Abstract]:Objective: To explore the protective effect of acupuncture on neuron cells and its possible molecular mechanism. Methods: intracerebroventricular injection of ibotenic acid to establish neuron injury model in rats; after the success of the model, given acupuncture and drug intervention, to observe the behavioral changes of rats, hippocampal neuron cell morphology; Determination of monoamine neurotransmitters (5-HT, NE), the content changes of brain-derived neurotrophic factor (BDNF) changes and apoptosis protein Bax/Bcl-2. Results: compared with the blank group, the model group showed a reduction in the amount of movement (P0.05), decreased appetite and ability to adapt to the new environment (P0.05), reduced the numbers of neurons, increased, nuclear vacuoles in loose connections, 5-HT and NE were significantly decreased (P0.05), Bax/Bcl-2 ratio (P0.05). Acupuncture and drug intervention, rats weight, appetite and ability to adapt to the new environment has been strengthened (P0.05), the increase in the number of neurons, loosely connected structure is improved, the number of nuclear vacuoles reduced, 5-HT, NE and BDNF were significantly increased (P0.05), Bax/Bcl-2 ratio (P0.05). Conclusion: the protective effect of acupuncture on neurons may be mediated by increasing the content of 5-HT, NE and BDNF and reducing neuronal cell apoptosis.
