續(xù)斷HPLC特征圖譜及3種活性成分的含量測定

【摘要】:目的:建立同時(shí)測定續(xù)斷中川續(xù)斷皂苷Ⅵ、馬錢苷、當(dāng)藥苷3種活性成分含量的方法,并建立續(xù)斷藥材HPLC特征圖譜。方法:采用Thermo BDS Hypersil C_(18)(250 mm×4.6 mm,5μm)色譜柱,以乙腈-0.1%磷酸溶液為流動(dòng)相進(jìn)行梯度洗脫,流速1.0 mL/min,柱溫30℃,檢測波長220 nm。結(jié)果:不同產(chǎn)地10批續(xù)斷樣品的HPLC譜圖具有很好的一致性,標(biāo)定了12個(gè)共有峰;地理位置相鄰的鄂西南、湘西北共有8批樣品的HPLC圖譜相似度大于0.94。各樣品的HPLC指紋圖譜相似度結(jié)果與3種活性成分總量聚類分析結(jié)果一致。結(jié)論:所建立的HPLC特征圖譜和含量測定方法能較好地評(píng)價(jià)和控制續(xù)斷藥材的質(zhì)量,從指紋圖譜化學(xué)成分信息的角度驗(yàn)證了包括五峰在內(nèi)的鄂西南(包括臨近的湘西北地區(qū))所產(chǎn)續(xù)斷的道地性或優(yōu)質(zhì)性。
