pH對DNA折紙納米結(jié)構(gòu)折疊與溶液穩(wěn)定性的影響

【摘要】:研究了不同pH環(huán)境對2種使用M13鏈作為骨架鏈的常見DNA折紙折疊與穩(wěn)定性的影響,獲得了其在pH=4~11這一廣泛范圍內(nèi)的耐受性數(shù)據(jù).首先觀察了在不同pH緩沖液中的三角形DNA折紙自組裝折疊行為,發(fā)現(xiàn)其合適pH范圍為6~9,過高或過低的pH都不能退火形成折紙結(jié)構(gòu).另一方面,又研究了在標(biāo)準(zhǔn)pH緩沖液中預(yù)先制備的DNA折紙,對不同pH環(huán)境的耐受性.研究發(fā)現(xiàn),在pH值為5~10的溶液環(huán)境中,DNA折紙均保持一定穩(wěn)定.比較以上2種研究方法獲得的結(jié)果,可以發(fā)現(xiàn),相比在不同pH緩沖液中從頭退火制備,已形成的DNA折紙對酸堿有較好耐受性.另外,研究也發(fā)現(xiàn),長方形折紙和三角形折紙穩(wěn)定性相似,都能在pH=5~10的環(huán)境中保持至少12 h的穩(wěn)定性.據(jù)此,還提出了pH對折紙穩(wěn)定性的可能影響機(jī)理,認(rèn)為其對氫鍵的破壞及對DNA鏈造成的斷裂共同導(dǎo)致了DNA折紙結(jié)構(gòu)的破碎.
[Abstract]:The effects of different pH environments on folding and stability of two kinds of common DNA origami with M13 chains as frame chains were studied, and their tolerability data in a wide range of pH=4~11 were obtained. First, the self-assembly and folding behavior of triangular DNA origami in different pH buffer was observed. It was found that the suitable pH range was 6? 9, and too high or too low pH could not anneal to form origami structure. On the other hand, the tolerance of DNA origami prepared in standard pH buffer to different pH environments was studied. It was found that the DNA origami kept certain stability in the solution environment of pH 5? 10. Comparing the results obtained by the above two methods, it can be found that the formed DNA origami is more tolerant to acid and base than that prepared in different pH buffers by ab initio annealing. In addition, it is found that the stability of rectangular origami is similar to that of triangular origami, and the stability of both rectangular origami and triangular origami can be kept at least 12 h in the environment of pH=5~10. Based on this, the possible mechanism of the influence of pH on the stability of origami paper is put forward. It is considered that the destruction of hydrogen bond and the fracture of DNA chain result in the fragmentation of DNA origami structure.
