HPLC法同時(shí)測(cè)定雪松不同部位5種酚酸類(lèi)成分的含量

【摘要】:目的:建立同時(shí)測(cè)定雪松中沒(méi)食子酸、原兒茶酸、兒茶素、咖啡酸和阿魏酸的含量測(cè)定方法,并用于雪松不同部位(松針、花序、嫩枝、木質(zhì)部、樹(shù)皮)該5種酚酸類(lèi)成分的測(cè)定。方法:采用Eclipse Plus-C18(250 mm×4.6 mm,5μm)色譜柱,以乙腈(A)-0.1%磷酸水溶液(B)為流動(dòng)相,梯度洗脫(0~6 min,7%A;6~70 min,7%A→20%A;70~80 min,20%A),流速1.0 m L·min~(-1),檢測(cè)波長(zhǎng):280 nm,柱溫:25℃。結(jié)果:沒(méi)食子酸、原兒茶酸、兒茶素、咖啡酸和阿魏酸質(zhì)量濃度分別在0.06~2.4、0.3~7.2、1.3~31.2、0.6~14.4和0.5~12μg·m L~(-1)范圍內(nèi)呈良好的線(xiàn)性關(guān)系,相關(guān)系數(shù)為0.999 6~0.999 9;平均加樣回收率(n=6)分別為93.4%、102.2%、100.3%、99.6%和98.6%;精密度、重復(fù)性和穩(wěn)定性良好,RSD均小于3%。雪松各部位中5種酚酸類(lèi)化合物的含量存在明顯差異,嫩枝中沒(méi)食子酸、兒茶素和阿魏酸的含量最高,分別為0.134、2.345和0.838 mg·g~(-1),松針中原兒茶酸的含量最高為0.479 mg·g~(-1),木質(zhì)中咖啡酸的含量最高1.113 mg·g~(-1),5種酚酸之和由高至低依次為嫩枝、木質(zhì)、樹(shù)皮、松針、花序。結(jié)論:經(jīng)方法學(xué)驗(yàn)證,本法可用于雪松不同部位中5種酚酸類(lèi)成分的含量測(cè)定。
