Slc11a2第六跨膜區(qū)在十二烷基磷酰膽堿膠束中的結(jié)構(gòu)

【摘要】:采用圓二色譜和核磁共振波譜研究了Slc11a2第六跨膜區(qū)在十二烷基磷酰膽堿(DPC)膜模擬環(huán)境中的結(jié)構(gòu)和定位.結(jié)果表明,Slc11a2第六跨膜區(qū)在DPC膠束中N端和C端各形成一小段α螺旋,兩段螺旋中間通過高度靈活的區(qū)域連接,整個(gè)肽插入到DPC膠束中.H267A突變使螺旋長(zhǎng)度變長(zhǎng),H272A突變使螺旋長(zhǎng)度略有變短,突變后結(jié)構(gòu)的變化可能和蛋白功能缺失相關(guān).
[Abstract]:The structure and localization of Slc11a2 sixth transmembrane region in the simulated environment of twelve alkyl phosphachcholine (DPC) membrane were studied by circular two chromatography and nuclear magnetic resonance spectroscopy. The results showed that the Slc11a2 sixth transmembrane region formed a small segment of alpha helix in the N and C ends of the DPC micelles, and the two segment helix was connected by a highly flexible region, and the whole peptide was inserted into DP. The mutation of.H267A in C micelles makes the length of helix longer. H272A mutation shortens the length of the helix, and the structural changes after mutation may be related to the loss of protein function.
