唾液及頭發(fā)中內(nèi)源性大麻素的高效液相色譜-串聯(lián)質(zhì)譜檢測

【摘要】:基于高效液相色譜-串聯(lián)質(zhì)譜儀建立了檢測唾液和頭發(fā)中花生四烯酸乙醇氨(AEA)和2-花生四烯酸甘油(2-AG)含量的方法,并用于檢測157名在校大學(xué)生唾液和頭發(fā)中的AEA和2-AG,探討AEA和2-AG對性別和肥胖的影響.唾液中的待測物采用乙酸乙酯液液萃取提純;頭發(fā)中的待測物經(jīng)甲醇孵育,用固相萃取柱萃取提純.在大氣壓化學(xué)電離正離子模式和多反應(yīng)監(jiān)測方式下進(jìn)行定性測試,以外標(biāo)法進(jìn)行定量測試.方法驗(yàn)證結(jié)果顯示,唾液AEA,2-AG檢測限分別為0.07和0.40 ng/m L;頭發(fā)AEA,2-AG檢測限分別為0.7和3.2 pg/mg;日內(nèi)、日間變異系數(shù)均小于15%,回收率為97.9%~108.0%.人群研究結(jié)果表明,肥胖者唾液和頭發(fā)AEA,2-AG含量均顯著高于非肥胖者(p0.010);男性唾液2-AG含量顯著低于女性(p0.050),但AEA含量沒有性別差異(p=0.844);男性頭發(fā)2-AG含量顯著高于女性(p0.010),但AEA含量沒有性別差異(p=0.456).
[Abstract]:Based on high performance liquid chromatography-tandem mass spectrometer, a method for the determination of arachidonic acid ethanolamine (AEAA) and 2-arachidonic acid glycerin (2-AGG) in saliva and hair was established.AEA and 2-AGG in saliva and hair of 157 college students were detected to investigate the effects of AEA and 2-AG on gender and obesity.The samples in saliva were extracted with ethyl acetate, and those in hair were incubated with methanol and extracted by solid phase extraction column.The qualitative test was carried out under the atmosphere pressure chemical ionization positive ion mode and the multi-reaction monitoring mode, and the quantitative test was carried out by external standard method.The results showed that the detection limit of AEAA 2-AG in saliva was 0.07 and 0.40 ng/m / L, the detection limit of AEAA 2-AG in hair was 0.7 and 3.2 PG / mg, and the coefficient of variation in day was less than 15 and the recovery rate was 108.0%.The results of the population study show thatThe content of AEAA 2-AG in saliva and hair of obese group was significantly higher than that in non-obese group (p0.010), the content of 2-AG in male saliva was significantly lower than that in female (P 0.050), but the content of AEA was not different in sex. The content of 2-AG in male hair was significantly higher than that in female (P 0.010 0), but the content of AEA in male was not different from that in female (P 0.456).
