HPLC測定決明不同組織中大黃酚和橙黃決明素含量

【摘要】:為了測定決明不同組織中大黃酚和橙黃決明素的含量。分別收集了決明種子、根、莖、花、葉、果莢、子葉、胚軸、愈傷與不定根等組織,利用回流提取法提取大黃酚與橙黃決明素,高效液相色譜法(HPLC)進行含量測定。采用依利特反相色譜柱,以大黃酚和橙黃決明素為指標(biāo),以乙腈-0.1%磷酸水為流動相進行梯度洗脫,流速1 m L/min,檢測波長為284 nm,柱溫30℃。結(jié)果表明:大黃素型蒽醌類化合物在決明中的分布具有明顯的組織特異性,其中,決明種子、胚軸、不定根、子葉與愈傷組織中含有大黃酚與橙黃決明素,決明根中僅含有大黃酚,其余組織不含有大黃酚與橙黃決明素,其余總蒽醌含量依次為子葉不定根愈傷組織胚軸根花。決明種子成熟過程中,大黃酚的含量呈現(xiàn)"波浪式上升"的趨勢,峰值出現(xiàn)在11月下旬;而橙黃決明素的含量呈現(xiàn)"先上升后下降"的趨勢,峰值出現(xiàn)在11月中旬。由于大黃酚與橙黃決明素的含量在決明不同成熟時期分別出現(xiàn)峰值,建議采收11月下旬的決明種子以獲得大黃酚,采收11月中上旬的決明種子以獲得較多的橙黃決明素。
[Abstract]:In order to determine the content of chrysophanol and orange cassia in different tissues of Cassia sinensis, seeds, roots, stems, flowers, leaves, fruit pods, cotyledons, cotyledons, cotyls, callus and adventitious roots were collected. Refluxing extraction method was used to extract chrysophanol and orange cassia. HPLC was used to determine the content of chrysophanol and orange cassia.
