牛油果中化學成分的內(nèi)部萃取電噴霧電離質(zhì)譜檢測

【摘要】:采用內(nèi)部萃取電噴霧電離質(zhì)譜(iEESI-MS)技術(shù),以甲醇作為萃取溶劑,無需樣品預處理,在優(yōu)化條件下分別對牛油果果肉和果皮中化學成分進行快速直接鑒定,并考察了不同成熟度牛油果果肉中化學成分的差異.實驗結(jié)果表明,在負離子檢測模式下,從果肉中鑒定出棕櫚酸、硬脂酸、棕櫚油酸、油酸和亞麻酸等12種有效營養(yǎng)成分,從果皮中鑒定出兒茶素、綠原酸等11種營養(yǎng)成分.利用正交偏最小二乘判別分析法(OPLS-DA)對不同成熟度的牛油果果肉的iEESI-MS指紋譜圖數(shù)據(jù)進行分析發(fā)現(xiàn),此方法能夠有效判別不同成熟程度的牛油果且5種化學成分差異顯著.本方法無需樣品預處理、樣品耗量少、分析速度快(單個樣品檢測時間小于1 min)且操作簡便,為植物有效營養(yǎng)成分和醫(yī)用價值的開發(fā)提供了一種快速質(zhì)譜分析新方法.
[Abstract]:The internal extraction electrospray ionization mass spectrometry (iEESI-MSS) technique was used to identify the chemical constituents of avocado pulp and pericarp with methanol as extraction solvent and without sample pretreatment. The results showed that palmitic acid, stearic acid, palmitic oil acid, oleic acid and linolenic acid were identified from the pulp under the negative ion detection mode. Eleven nutritive components, such as catechin and Lv Yuan acid, were identified from pericarp. The iEESI-MS fingerprint data of avocado pulp with different maturity were analyzed by orthogonal partial least squares discriminant analysis (OPLS-DA). This method can effectively distinguish avocados of different maturation degree and the difference of five chemical components is significant. This method does not need the sample pretreatment, the sample consumption is less, the analysis speed is fast (the detection time of a single sample is less than 1 min), and the operation is simple and convenient. A new rapid mass spectrometry analysis method is provided for the development of plant effective nutrients and medical value.
