不同蜂針劑量對類風(fēng)濕性關(guān)節(jié)炎大鼠成纖維樣滑膜細(xì)胞活力的影響

【摘要】:目的:觀察不同劑量的蜂針對佐劑誘導(dǎo)的類風(fēng)濕性關(guān)節(jié)炎大鼠成纖維滑膜細(xì)胞活力的影響,探討蜂針治療類風(fēng)濕性關(guān)節(jié)炎的可能機(jī)制。方法:建立大鼠佐劑性類風(fēng)濕性關(guān)節(jié)炎動物模型,將Wistar大鼠隨機(jī)分為空白組、模型組和單蜂1 min組、雙蜂1 min組、三蜂1 min組、單蜂30 s組,觀察各組足底厚度以及HE染色的病理變化,同時(shí)關(guān)節(jié)滑膜取材,采用MTT法檢測成纖維樣滑膜細(xì)胞增殖情況。結(jié)果:足底厚度比較,1 min蜂針組較模型組以及其他蜂針劑量組別低(P0.01);HE病理形態(tài)學(xué)評分,模型組病理形態(tài)學(xué)改變評分較各蜂針治療組高(P0.05);MTT檢測成纖維滑膜細(xì)胞活力中,24 h時(shí)間點(diǎn)開始出現(xiàn),1 min蜂針組抑制成纖維細(xì)胞增殖較模型組顯著(P0.05),48 h較24 h時(shí)間點(diǎn)各組差異更顯著。結(jié)論:適當(dāng)劑量的蜂針能減輕類風(fēng)濕性關(guān)節(jié)炎大鼠足底厚度,降低病理形態(tài)學(xué)評分,其機(jī)制可能跟蜂針抑制成纖維樣滑膜細(xì)胞增殖活力有關(guān)。
[Abstract]:Aim: to observe the effects of different doses of bee on the viability of fibroblast synoviocytes induced by adjuvant in rats with rheumatoid arthritis (RA), and to explore the possible mechanism of bee acupuncture in treating rheumatoid arthritis (RA). Methods: animal models of adjuvant rheumatoid arthritis were established in rats. Wistar rats were randomly divided into blank group, model group and single bee 1 min group, double bee 1 min group, three bees 1 min group and single bee 30 s group. The thickness of plantar and the pathological changes of HE staining were observed. Meanwhile, the synovial membrane of the joint was taken and the proliferation of fibroid synovial cells was detected by MTT method. Results: compared with the plantar thickness, 1 min bee needle group was lower than the model group and other bee needle dose groups (P0.01); HE pathomorphology score, the model group was higher than each bee needle treatment group (P0.05); The proliferation of fibroblasts was inhibited by MTT at 24 h, and the inhibition of fibroblast proliferation in min group was significantly higher than that in model group (P0.05), and the difference at 48 h was more significant than that at 24 h. Conclusion: the appropriate dose of bee acupuncture can reduce the plantar thickness and the pathological morphology score of rheumatoid arthritis rats. The mechanism may be related to the inhibition of fibroid synovial cell proliferation by bee needle.
