不同生長期采集的蒙藥森登化學(xué)成分比較研究

【摘要】:目的:該文照高效液相色譜法(HPLC)同時測定32批蒙藥材森登中表兒茶素、二氫槲皮素、楊梅素3個成份的含量測定及建立森登HPLC指紋圖譜,并進(jìn)行單體分離實(shí)驗(yàn),為森登的質(zhì)量控制及繼續(xù)研究提供依據(jù)。方法:含量測定及指紋圖譜研究:該方法色譜條件為ODS C18色譜柱(4.6×300mm,5μm),以乙腈(B)和0.1%的醋酸(A)為流動相,檢測波長為274nm,梯度洗脫,流速1mL/min,進(jìn)樣量5μL,柱溫30℃。單體分離研究:取森登的粗粉,加95%的乙醇回流提取3次,低溫(40℃)回收乙醇濃縮加少量蒸餾水,依次加同體積石油醚、二氯甲烷、正丁醇萃取,取3個萃取液。二氯甲烷萃取液凝膠柱色譜進(jìn)行分離。對分離得到的化合物進(jìn)行光譜結(jié)構(gòu)鑒定。結(jié)果:含量測定中顯示表兒茶素、二氫槲皮素、楊梅素3個被測組分達(dá)到基線分離;在線性范圍內(nèi)具有良好的線性關(guān)系,相關(guān)系數(shù)均大于0.999;平均回收率分別為99.61%、99.73%、99.91%。不同來源的27批森登進(jìn)行HPLC指紋圖譜研究,與共有模式之間具有良好的相似性,相似度大于0.99。HPLC指紋圖譜研究中發(fā)現(xiàn)23種共有峰,其中表兒茶素、二氫槲皮素、二氫楊梅素、楊梅素、槲皮素、柑橘素6個成份與標(biāo)準(zhǔn)品對照已確定。單體分離實(shí)驗(yàn)中,分離出兒茶素、二氫楊梅素、1-O-甲基-D肌-肌醇3個化合物,其中三號化合物為森登里首次分離得到的新化合物。結(jié)論:該研究不僅作為蒙藥森登質(zhì)量控制的基礎(chǔ),還進(jìn)一步道地藥材的采收,加工、有效成分的分析開發(fā)利用提供研究基礎(chǔ)。
