電針對腦缺血大鼠丘腦腹后外側(cè)核神經(jīng)生長因子和生長休止特定蛋白7表達的影響
[Abstract]:Objective: to observe the effect of electroacupuncture on the expression of (VPL) nerve growth factor (NGF) and growth rest specific protein 7 (Gas 7) in ventrolateral nucleus of thalamus of rats with focal cerebral ischemia. To explore the possible mechanism of electroacupuncture in treating secondary injury of focal cerebral ischemia. Methods: 48 SD rats were randomly divided into normal group, sham operation group, model group and electroacupuncture group after ischemia (12 rats in each group). The right middle cerebral artery (MCA) embolization model was established by thread embolization. In the electroacupuncture group, the points of Baihui and Zusanli on the left side were treated once a day for 30 min, for 21 days. The expression of NGF and Gas 7 in the right VPL of rats was detected by immunohistochemistry and Western blot, and the morphology of VPL neurons was observed by Nissl staining. Results: the results of Nissl staining showed that the right VPL structure of normal group and sham operation group was clear and complete, the nucleus was in the middle, the nucleolus was clear, the VPL neurons in the model group were stained deeply and the nucleus was pyknosis, and the morphology of nerve cells in the electroacupuncture group was close to that in the normal group. The expression of NGF and Gas 7 protein in VPL of sham-operated group was not significantly different from that of normal group (P0.05). Compared with the normal group, the expression of NGF and Gas 7 protein in VPL of the model group was significantly higher than that in the model group (P0.05); compared with the model group, the expression of NGF and Gas 7 protein in the VPL of the electroacupuncture group was significantly higher (P0.05). Conclusion: the increased expression of NGF and Gas 7 in VPL of rats with focal cerebral ischemia may be involved in the process of compensatory recovery. Electroacupuncture can up-regulate the expression of NGF and Gas 7 in VPL of ischemic side, which may be one of the mechanisms of electroacupuncture in ameliorating the secondary thalamic damage after cerebral ischemia.
