【摘要】：目的通過建立大鼠類風(fēng)濕關(guān)節(jié)炎模型,予以壯醫(yī)藥線點(diǎn)灸干預(yù),觀察治療效果,并進(jìn)一步探討其對大鼠關(guān)節(jié)滑膜細(xì)胞凋亡的影響。方法建立佐劑關(guān)節(jié)炎大鼠模型,予以甲氨蝶呤(MTX組)、壯醫(yī)藥線點(diǎn)灸(ZHM組)和兩者聯(lián)合(CT組)干預(yù)。觀察并評估大鼠關(guān)節(jié)炎指數(shù)和腫脹度;HE染色觀察關(guān)節(jié)滑膜病理變化;TUNEL染色觀察大鼠關(guān)節(jié)滑膜細(xì)胞凋亡率;Western blotting檢測滑膜組織中Bcl-2/Bax的表達(dá)。結(jié)果 MTX組(P0.05)、ZHM組和CT組(P0.05)大鼠關(guān)節(jié)炎系數(shù)相比于關(guān)節(jié)炎模型組(RA組)均有不同程度的降低,其中CT組降低最為顯著,ZHM組與MTX組相比差異亦具統(tǒng)計學(xué)意義(P0.05);HE染色結(jié)果顯示MTX組、ZHM組和CT組大鼠滑膜襯里層細(xì)胞增生和炎性細(xì)胞浸潤與RA組相比均有不同程度的改善;MTX組(P0.05)、ZHM組(P0.05)和CT組(P0.05)與RA組相比細(xì)胞凋亡數(shù)量顯著增加,差異具有統(tǒng)計學(xué)意義,CT組和ZHM組與MTX組比較,細(xì)胞凋亡現(xiàn)象顯著,差異具有統(tǒng)計學(xué)意義(P0.05);MTX組(P0.05)、ZHM組(P0.05)和CT組(P0.05)與RA組相比大鼠關(guān)節(jié)滑膜組織中Bcl-2蛋白表達(dá)顯著下調(diào),Bax顯著上調(diào),CT組和ZHM組與MTX組相比Bcl-2下調(diào)、Bax上調(diào)均更為顯著,差異具統(tǒng)計學(xué)意義(P0.05)。結(jié)論壯醫(yī)藥線點(diǎn)灸干預(yù)能夠在一定程度上改善佐劑關(guān)節(jié)炎大鼠癥狀,聯(lián)合甲氨蝶呤使用效果更為顯著;而且其作用機(jī)制可能是通過Bcl-2信號通路調(diào)控關(guān)節(jié)滑膜細(xì)胞凋亡來實(shí)現(xiàn)的。
[Abstract]:Objective to establish a rat model of rheumatoid arthritis (RA) and to observe the therapeutic effect of moxibustion on synovial cells in rats. Methods the adjuvant arthritis model was established in rats. Methotrexate group (MTX group), Zhuang medicine thread moxibustion group (ZHM group) and combined therapy group (CT group) were given intervention. The index of arthritis and degree of swelling were observed and evaluated; the pathological changes of synovial membrane were observed by HE staining; and the expression of Bcl-2/Bax in synovial tissue was detected by; Western blotting by TUNEL staining. Results the arthritis coefficient in MTX group (P0.05), ZHM group) and CT group (P0.05) was lower than that in model group (RA group). The decrease in CT group was the most significant, and the difference between ZHM group and MTX group was statistically significant (P0.05); HE staining results showed that the proliferation of synovial lining cells and inflammatory cell infiltration in MTX group, ZHM group and CT group were different from RA group). The number of apoptosis in MTX group (P0.05), ZHM group) and CT group (P0.05) was significantly higher than that in RA group. The difference was statistically significant. Compared with MTX group, apoptosis in CT group and ZHM group was significant. The difference was statistically significant (P0.05); MTX group (P0.05), ZHM group, P0.05) and CT group (P0.05) compared with the RA group, the expression of Bcl-2 protein significantly decreased, Bax significantly up-regulated, CT group and ZHM group compared with MTX group Bcl-2 down-regulation, Bax up-regulation was more significant, the difference was statistically significant (P0.05). Conclusion the intervention of Zhuang medicine thread moxibustion can improve the symptoms of adjuvant arthritis rats to a certain extent, and the effect of combined use of methotrexate is more remarkable, and its mechanism may be realized by regulating the apoptosis of synovial cells through Bcl-2 signaling pathway.
