民族藥野蠶豆根抗心肌缺血和抗凝血作用研究

【摘要】:目的觀察民族藥野蠶豆根抗心肌缺血和抗凝血等心血管方面相關(guān)的藥理活性,為進(jìn)一步探究野蠶豆根的臨床應(yīng)用方向和開(kāi)發(fā)野蠶豆根的藥用價(jià)值提供依據(jù)。方法對(duì)野蠶豆根進(jìn)行急性毒性評(píng)價(jià)后,采用毛細(xì)玻管法、小鼠斷尾法以及血漿復(fù)鈣試驗(yàn)探究野蠶豆根相關(guān)抗凝血活性;采用鹽酸異丙腎上腺素誘導(dǎo)小鼠急性心肌缺血模型研究野蠶豆根對(duì)心肌缺血損傷的保護(hù)作用;采用角叉菜膠致小鼠氣囊滑膜炎模型觀察野蠶豆根的抗炎活性。結(jié)果野蠶豆根用藥安全,能明顯增加小鼠的凝血時(shí)間和出血時(shí)間,其水、甲醇和乙酸乙酯提取部位也均能顯著延長(zhǎng)體外兔血漿復(fù)鈣時(shí)間,其中水提取部位時(shí)間延長(zhǎng)最明顯;且其能夠顯著降低小鼠急性心肌缺血模型中血清乳酸脫氫酶和丙二醛的含量,減少炎癥模型中前列腺素E2的滲出。結(jié)論野蠶豆根在抗心肌缺血和抗凝血等心血管相關(guān)方面均表現(xiàn)出了一定的藥理活性,值得對(duì)野蠶豆根在心血管方面的藥效機(jī)制和臨床應(yīng)用作進(jìn)一步的研究開(kāi)發(fā)。
[Abstract]:Objective to observe the pharmacological activities of Vicia faba root in the cardiovascular aspects such as myocardial ischemia and anticoagulant, so as to provide the basis for further exploring the clinical application direction of wild broad bean root and the medicinal value of developing wild broad bean root. Methods the acute toxicity evaluation of Vicia faba root was carried out, and the relative anticoagulant activity of Vicia faba root was investigated by capillary tube method, mouse tail cut method and plasma recalcium test. The protective effect of Vicia faba root on myocardial ischemia injury was studied in mice induced by isoproterenol hydrochloride, and the anti-inflammatory activity of Vicia faba root was observed by carrageenin induced mouse balloon synovitis model. Results the drug of Vicia faba root was safe and could significantly increase the blood coagulation time and bleeding time of mice. The water, methanol and ethyl acetate extracts could also significantly prolong the recalcium time of rabbit plasma in vitro, among which the time of water extraction was the most obvious. It can significantly reduce the contents of serum lactate dehydrogenase and malondialdehyde in mice with acute myocardial ischemia and decrease the exudation of prostaglandin E _ 2 in inflammatory model. Conclusion Vicia faba root has some pharmacological activities in cardiovascular related aspects such as myocardial ischemia and anticoagulant. It is worthy of further research and development of the mechanism and clinical application of Vicia faba root in cardiovascular system.
