針刺對(duì)高脂飲食誘導(dǎo)的肥胖小鼠白色脂肪組織巨噬細(xì)胞極化的影響

【摘要】:目的:觀察穴位與非穴位針刺對(duì)高脂飲食誘導(dǎo)的肥胖小鼠白色脂肪組織巨噬細(xì)胞極化的影響。方法:將50只雄性C 57 BL/6小鼠隨機(jī)抽取10只作為正常組,其余40只給予持續(xù)高脂飲食造模16周,將30只造模成功的小鼠隨機(jī)分為模型組、非穴位組、穴位組,每組10只,繼續(xù)給予高脂飲食喂養(yǎng)8周。正常組繼續(xù)給予普通飼料喂養(yǎng)8周。自造模成功后第2天開始,穴位組小鼠針刺"關(guān)元""足三里""胃脘下俞",非穴位組針刺尾部2個(gè)非穴位,每日1次,每次針刺15 min,連續(xù)治療8周。于造模成功后及末次治療后第2天記錄各組小鼠體質(zhì)量;眼眶采血,檢測小鼠血清三酰甘油(triglyceride,TG)、血清總膽固醇(total cholesterol,TC)水平;HE染色觀察各組小鼠附睪白色脂肪組織形態(tài),real-time quantitative PCR(RT-q PCR)檢測小鼠附睪白色脂肪中白細(xì)胞介素6(interleukin-6,IL-6)、單核細(xì)胞趨化蛋白-1(monocyte chemotactic protein 1,MCP-1)、腫瘤壞死因子α(tumor necrosis factorα,TNF-α)、白細(xì)胞介素10(interleukin-10,IL-10)、誘導(dǎo)型一氧化氮合酶(inducible nitric oxide synthesis,i NOS)和CD 206 m RNA表達(dá)情況,免疫組化(IHC)檢測附睪白色脂肪組織中i NOS和CD 206的蛋白表達(dá)情況。結(jié)果:與正常組比較,模型組、非穴位組、穴位組小鼠在喂養(yǎng)第16周和第24周時(shí)體質(zhì)量均明顯上升(均P0.05);與模型組比較,穴位組小鼠在第24周時(shí)體質(zhì)量明顯降低(P0.05)。與正常組比較,模型組TG、TC明顯上升(均P0.05);與模型組比較,非穴位組TC明顯降低(P0.05),穴位組TG、TC明顯降低(均P0.05)。與正常組比較,模型組和非穴位組小鼠脂肪組織中MCP-1、TNF-α、IL-6、i NOS m RNA的表達(dá)明顯升高(均P0.05),IL-10、CD 206 m RNA的表達(dá)明顯降低(均P0.05);與模型組比較,穴位組脂肪組織中MCP-1、TNF-α、IL-6、i NOS m RNA的表達(dá)明顯降低(均P0.05),IL-10、CD 206 m RNA的表達(dá)明顯升高(均P0.05)。HE染色可見:正常組脂肪組織呈蜂窩狀結(jié)構(gòu),由大量單泡脂肪細(xì)胞構(gòu)成,脂肪細(xì)胞呈多邊形或圓形,呈空泡狀;模型組脂肪細(xì)胞明顯增大且脂肪細(xì)胞大小不規(guī)則,細(xì)胞間隙變大;穴位組脂肪細(xì)胞變小,細(xì)胞間隙變小。與正常組比較,模型組和非穴位組小鼠脂肪組織中i NOS蛋白表達(dá)明顯升高(均P0.05),CD206蛋白表達(dá)明顯降低(均P0.05);與模型組比較,穴位組脂肪組織中i NOS蛋白表達(dá)明顯降低(P0.05),CD206蛋白表達(dá)明顯升高(P0.05)。結(jié)論:針刺"關(guān)元""足三里""胃脘下俞"穴可通過影響白色脂肪組織巨噬細(xì)胞極化改善肥胖小鼠脂肪炎性反應(yīng)。
[Abstract]:Aim: to observe the effect of acupuncture at acupoints and non-acupoints on the polarization of macrophages in white adipose tissue of obese mice induced by high fat diet. Methods: ten male C57BL / 6 mice were randomly selected as normal group, the other 40 mice were given continuous high-fat diet for 16 weeks, and 30 successful mice were randomly divided into model group (n = 10), non-acupoint group (n = 10) and control group (n = 10). Continue feeding on a high fat diet for 8 weeks. The normal group was given normal feed for 8 weeks. Since the second day after the successful establishment of the model, acupuncture of "Guanyuan" Zusanli "Weiwan Xiazu" in the acupoint group and 2 non-acupoints in the non-acupoint group were treated continuously for 8 weeks, once a day for 15 mins each time. The body mass of each group was recorded on the second day after the successful establishment of the model and the second day after the last treatment. Detection of serum triglyceride (TGG) and total cholesterol (TC) in mice by HE staining to observe the morphology of white adipose tissue in the epididymis of each group and to detect interleukin-6 interleukin-6mil and monocyte chemoattractant protein -1 monocyte in white fat of mouse epididymis The expression of TNF- 偽, IL-10, inducible nitric oxide synthase synthesis-inducible nitric oxide synthesis-iNOS) and CD206mRNA were detected by chemotactic protein 1, tumor necrosis factor 偽, TNF- 偽, interleukin-10, inducible nitric oxide synthesizer and CD206mRNA, respectively. The expression of iNOS and CD206 in white adipose tissue of epididymis was detected by immunohistochemistry (IHC). Results: compared with the normal group, the body weight of the model group, the non-point group and the acupoint group increased significantly at the 16th and 24th week of feeding (all P 0.05), and the body mass of the acupoint group decreased significantly at the 24th week compared with the model group. Compared with the normal group, the TGG TC in the model group was significantly higher than that in the model group (all P0.05A), and that in the non-acupoint group was significantly lower than that in the non-acupoint group, and in the acupoint group, the TC in the acupoint group was significantly lower than that in the model group (all P 0.05). Compared with the normal group, the expression of NOS mRNA in the adipose tissue of the model group and non-acupoint group was significantly higher than that in the model group (P0.05, P 0.05) and the expression of CD206 mRNA was significantly lower in the model group than that in the model group, and compared with the model group, the expression of IL-10 + CD206 mRNA in the model group was significantly lower than that in the control group. In adipose tissue of acupoint group, the expression of MCP-1TNF- 偽 IL-6i NOS mRNA was significantly decreased (P0.05 + IL-10 + CD206 mRNA expression was significantly increased (P0.05). He staining showed that normal adipose tissue was honeycombed and consisted of a large number of single follicular adipocytes. The adipocytes were polygonal or round and vacuolated; the adipocytes in the model group were obviously enlarged and the size of the adipocytes was irregular and the gap between the adipocytes became larger; in the acupoint group the adipocytes became smaller and the intercellular space became smaller. Compared with the normal group, the expression of iNOS protein in the adipose tissue of the model group and the non-acupoint group was significantly increased (P 0.05) and the expression of CD206 was significantly decreased (all P 0.05), and compared with the model group, the expression of iNOS was significantly higher than that of the control group. The expression of iNOS protein in adipose tissue of acupoint group decreased significantly. Conclusion: acupuncture at "Guanyuan", "Zusanli" and "Weiwan Xiashu" can improve the fat inflammatory response of obese mice by affecting the polarization of macrophages in white adipose tissue.
