不同刺灸法對(duì)大鼠功能性便秘干預(yù)作用的比較研究

【摘要】:目的:比較毫針刺法、電針和艾灸對(duì)大鼠功能性便秘的干預(yù)作用。方法:將60只雄性SD大鼠按隨機(jī)數(shù)字表法分為對(duì)照組(8只)、模型組(11只)、藥物組(8只)、毫針組(11只)、電針組(11只)和艾灸組(11只)。除對(duì)照組外,余組均以鹽酸洛哌丁胺混懸液連續(xù)灌胃6d制備功能性便秘模型,同時(shí)對(duì)照組以等體積飲用水灌胃。毫針組、電針組和艾灸組均于每日灌胃后1h分別給予毫針針刺、電針干預(yù)、艾條溫和灸,均取"天樞""上巨虛";藥物組予西沙必利混懸液灌胃,均每天干預(yù)1次,連續(xù)6d。計(jì)量各組大鼠干預(yù)最后1日24h攝食量、糞便量及糞便含水量,采用炭餐指示劑進(jìn)行腸推進(jìn)率試驗(yàn),應(yīng)用蘇木精-伊紅染色和阿利新藍(lán)染色觀察結(jié)腸組織結(jié)構(gòu)及黏膜層酸性黏液,采用實(shí)時(shí)熒光定量PCR(real-time PCR)法檢測(cè)結(jié)腸組織干細(xì)胞因子(stem cell factor,SCF)、c-kit mRNA的表達(dá)。結(jié)果:與對(duì)照組比較,模型組24h攝食量、糞便量均減少(均P0.01),糞便含水量、腸推進(jìn)率均降低(均P0.01);與模型組比較,藥物組、毫針組、電針組24h糞便量、糞便含水量均增加(P0.05,P0.01),艾灸組差異無統(tǒng)計(jì)學(xué)意義(P0.05),各干預(yù)組腸推進(jìn)率均升高(均P0.01);與藥物組比較,毫針組、電針組和艾灸組24h攝食量、糞便量差異均無統(tǒng)計(jì)學(xué)意義(均P0.05),糞便含水量僅艾灸組降低(P0.01),毫針組、艾灸組腸推進(jìn)率均低于藥物組(均P0.01),而電針組與藥物組差異無統(tǒng)計(jì)學(xué)意義(P0.05);艾灸組糞便含水量低于毫針組、電針組(均P0.01),毫針組、艾灸組腸推進(jìn)率均低于電針組(均P0.01)。HE染色顯示各組大鼠結(jié)腸組織結(jié)構(gòu)正常、完整,各組間無明顯差異。阿利新藍(lán)染色顯示黏膜層黏液物質(zhì),與對(duì)照組比較,模型組酸性黏液明顯減少;與模型組比較,藥物組、毫針組、電針組和艾灸組均有不同程度增加。與對(duì)照組比較,模型組結(jié)腸組織SCF、c-kit mRNA表達(dá)均降低(均P0.05);與模型組比較,藥物組、毫針組、電針組和艾灸組SCF、c-kit mRNA表達(dá)均升高(均P0.05);毫針組、電針組c-kit mRNA表達(dá)均低于艾灸組(均P0.05)。結(jié)論:毫針刺法、電針和艾灸均對(duì)大鼠功能性便秘具有不同程度的良性調(diào)整作用,以電針作用最佳,毫針刺法次之。
[Abstract]:Objective: to compare the interventional effects of Milliform Acupuncture, electroacupuncture and moxibustion on functional constipation in rats. Methods: sixty male Sprague-Dawley rats were randomly divided into control group (n = 8), model group (n = 11), drug group (n = 8), filiform group (n = 11), electric acupuncture group (n = 11) and moxibustion group (n = 11). Except the control group, the functional constipation model was established by continuous oral administration of loperamide hydrochloride suspension for 6 days, while the control group was perfused with the same volume of drinking water. The patients in the filiform acupuncture group, electroacupuncture group and moxibustion group were treated with filiform acupuncture, electroacupuncture intervention, moxa mild moxibustion, "Tianshu" and "Shangzuxu", respectively, and the drug group were given cisapride suspension once a day for 6 days. The feeding amount, fecal quantity and fecal water content of rats in each group were measured at 24 h after intervention on the last day. The intestinal propulsion rate was tested with charcoal meal indicator. The colonic tissue structure and mucous mucus in mucous membrane were observed by hematoxylin eosin staining and alimine blue staining. The expression of stem cell factor (SCFK) c-kit mRNA in colon tissue was detected by real-time fluorescence quantitative PCR(real-time. Results: compared with the control group, 24 h food intake and fecal quantity in the model group were decreased (all P 0.01), the fecal water content and intestinal propulsive rate were all decreased (all P 0.01), and compared with the model group, 24 h fecal quantity was decreased in the drug group, the filiform needle group and the electroacupuncture group. The fecal water content was increased in both groups, but there was no significant difference between the moxibustion group and the moxibustion group, but the intestinal propulsive rate in each intervention group was all increased (all P 0.01), compared with the drug group, the intakes of the millimeter needle group, the electroacupuncture group and the moxibustion group were 24 h, and that of the moxibustion group was higher than that of the control group. There was no significant difference in fecal quantity (P 0.05). The fecal water content in moxibustion group was lower than that in moxibustion group (P 0.01), while that in the filiform needle group was lower than that in the control group. The intestinal propulsive rate of moxibustion group was lower than that of drug group (all P 0.01), but there was no significant difference between electroacupuncture group and drug group, while the fecal water content in moxibustion group was lower than that in millineedle group, and that in electroacupuncture group was lower than that in millineedle group. The intestinal propulsive rate of moxibustion group was lower than that of electroacupuncture group (P0.01).HE staining showed that the colonic tissue structure of each group was normal and intact, and there was no significant difference between each group. Compared with the control group, the acid mucus in the model group decreased significantly, and the drug group, the millimeter needle group, the electroacupuncture group and the moxibustion group increased in varying degrees compared with the model group. Compared with the control group, the expression of SCFc-kit mRNA in colonic tissue of model group was lower than that in control group (P0.05), and the expression of SCFc-kit mRNA in drug group, millimeter acupuncture group, electroacupuncture group and moxibustion group was higher than that in model group (all P 0.05), and the expression of c-kit mRNA in acupuncture group and electroacupuncture group was lower than that in moxibustion group (P 0.05). Conclusion: the effect of acupuncture, electroacupuncture and moxibustion on functional constipation in rats was better than that of acupuncture.
