電針對去卵巢大鼠骨形成及骨吸收活性的影響

【摘要】:目的研究電針刺對去卵巢骨質(zhì)疏松模型大鼠的全身骨密度(BMD)、骨礦物含量(BMC)及其血清對體外培養(yǎng)成骨細(xì)胞的骨形成因子骨鈣素(OPG)和骨吸收因子(RANKL)蛋白表達(dá)的影響。方法選取SPF級2月齡SD雌性大鼠40只,隨機(jī)分為空白組、電針組、藥物組和模型組,每組10只。其中空白組不做任何處理,后3組采用手術(shù)方法切除雙側(cè)卵巢。飼養(yǎng)3個(gè)月后,骨密度檢測結(jié)果顯示造模成功。隨后藥物組大鼠灌胃服用戊酸雌二醇3個(gè)療程,電針組大鼠采用針刺后通電治療3個(gè)療程并服用藥物組相同體積的蒸餾水,模型組服用相應(yīng)體積的蒸餾水,空白組常規(guī)飼養(yǎng)。各組大鼠于SPF級實(shí)驗(yàn)室處理3個(gè)月后,雙能X射線骨密度儀檢測全身骨密度和骨礦含量。隨后,水合氯醛麻醉各組大鼠,心臟采血,各組血液分別經(jīng)濾網(wǎng)過濾、56℃水浴處理后加入體外培養(yǎng)的大鼠成骨細(xì)胞培養(yǎng)液中,處理3d后多聚甲醛固定成骨細(xì)胞,采用免疫細(xì)胞化學(xué)染色法檢測成骨細(xì)胞內(nèi)OPG、RANKL蛋白的表達(dá)量。結(jié)果與空白組相比,模型組大鼠BMD,BMC顯著降低(P0.05),服用藥物或針刺處理后,BMD和BMC顯著增高(P0.05),與模型組相比差異具有統(tǒng)計(jì)學(xué)意義。各組大鼠血清處理成骨細(xì)胞后,模型組骨形成指標(biāo)OPG蛋白表達(dá)量較低,經(jīng)藥物或針刺后,OPG蛋白表達(dá)量顯著升高(P0.01)。此外,模型組骨吸收指標(biāo)RANKL的表達(dá)量較高,經(jīng)藥物和針刺處理后,RANKL蛋白表達(dá)量顯著下降(P0.01)。結(jié)論針刺能有效升高大鼠BMD和BMC,同時(shí)針刺處理后的大鼠血清具有很好的促進(jìn)骨形成和抑制骨吸收的能力。
[Abstract]:Objective to study the effects of electroacupuncture (EA) on the expression of bone morphogenetic factor osteocalcin (OPG) and bone resorption factor (RANKL) in osteoblasts of ovariectomized rats with bone mineral content (BMC) and bone mineral content (BMC).Methods Forty SPF 2-month-old SD female rats were randomly divided into blank group, electroacupuncture group, drug group and model group with 10 rats in each group.The blank group did not do any treatment, and the latter 3 groups underwent bilateral ovary resection.After feeding for 3 months, bone mineral density test showed that the model was successful.Then the rats in the drug group were treated with estradiol valerate for 3 courses, the rats in the electroacupuncture group were treated with electricity after acupuncture for 3 courses and the same volume of distilled water was taken in the drug group, the model group was given the corresponding volume of distilled water, and the blank group was fed routinely.The bone mineral density and bone mineral content were measured by dual energy X ray absorptiometry after 3 months of SPF laboratory treatment.Then, the rats were anesthetized by chloral hydrate, and the blood was collected from the heart.
