膝痹膏摩法對兔膝骨性關(guān)節(jié)炎滑膜MMP-1、MMP-3、MMP-13影響的研究

【摘要】:目的:通過觀察膝痹膏摩法對兔膝骨性關(guān)節(jié)炎滑膜中MMP-1、MMP-3、MMP-13水平的影響,對探究膝痹膏摩法治療膝骨性關(guān)節(jié)炎的作用機(jī)制提供實驗依據(jù)。方法:新西蘭大白兔43只,隨機(jī)分出10只作為正常組,將剩余33只利用石膏鐵絲固定法建立兔膝骨性關(guān)節(jié)炎模型,造模8周,隨機(jī)處死3只兔子查看造模情況,造模成功,將30只兔子隨機(jī)分成模型組、手法組、膝痹膏摩組,每組10只,正常組和模型組正常飼養(yǎng),不做干預(yù);手法組給予推拿手法治療每天一次,治療8周;膝痹膏摩組給予膝痹膏配合推拿手法治療每天一次,治療8周。治療結(jié)束后取材,通過大體觀察關(guān)節(jié)面和關(guān)節(jié)軟骨行HE病理染色觀察軟骨表面組織學(xué)形態(tài)變化情況;滑膜采用雙抗體夾心酶聯(lián)免疫吸附法(ELISA)檢測各組滑膜組織中MMP-1、MMP-3、MMP-13的表達(dá)情況。結(jié)果:大體觀察及光鏡下觀察軟骨表面:與模型組相比,手法組和膝痹膏摩組軟骨損傷程度較輕,其中膝痹膏摩組損傷程度最輕,大體評分膝痹膏摩組和模型組比較有統(tǒng)計學(xué)意義(p0.05)。膝關(guān)節(jié)軟骨Mankin評分:模型組膝關(guān)節(jié)軟骨Mankin評分顯著高于其他各組(p0.01);而手法組及膝痹膏摩組與模型組比較膝關(guān)節(jié)軟骨Mankin評分顯著降低(p0.01),其中膝痹膏摩組膝關(guān)節(jié)軟骨Mankin評分較手法組更低(p0.05)。結(jié)果顯示模型組中MMP-1、MMP-3、MMP-13含量均高于其余各組(P0.01),正常組中MMP-1、MMP-3、MMP-13含量最低(P0.01)。各組間比較:膝痹膏摩組MMP-13的含量明顯低于手法組(P0.01);膝痹膏摩組MMP-1、MMP-3的含量和手法組比較有統(tǒng)計學(xué)意義(P0.05)。結(jié)論:膝痹膏摩法治療膝骨性關(guān)節(jié)炎可能是通過抑制MMP-1、MMP-3、MMP-13酶的活性而延緩對軟骨及滑膜的破壞,從而減慢對關(guān)節(jié)組織的損害程度,發(fā)揮治療膝骨性關(guān)節(jié)炎的作用。
[Abstract]:Objective: to observe the effect of knee arthralgia on the level of MMP-1, MMP-3 and MMP-13 in the synovium of rabbit knee osteoarthritis, and to provide experimental basis for exploring the mechanism of knee arthritic arthritis treated by knee arthralgia.Methods: 43 New Zealand white rabbits were randomly divided into 10 normal groups. The remaining 33 rabbits were used to establish the rabbit knee osteoarthritis model by the method of plaster wire fixation.30 rabbits were randomly divided into model group, manipulation group, knee arthralgia group with 10 rabbits in each group, normal feeding group and model group without intervention, manipulation group was treated with massage manipulation once a day for 8 weeks.The treatment group was treated with knee arthralgia ointment and massage manipulation once a day for 8 weeks.After treatment, histopathological changes of articular surface and articular cartilage were observed by HE pathological staining, and the expression of MMP-1, MMP-3 and MMP-13 in synovial tissue were detected by double antibody sandwich enzyme linked immunosorbent assay (Elisa).Results: compared with the model group, the degree of cartilage injury was lighter in manipulation group and knee arthralgia group, especially in knee arthralgia group.The gross score of the two groups was significantly higher than that of the model group (P 0.05).The Mankin score was lower than that in the manipulation group (P 0.05).The results showed that the content of MMP-1, MMP-3, MMP-13 in the model group was higher than that in the other groups, and the content of MMP-1, MMP-3, MMP-13 in the normal group was the lowest (P 0.01).Comparison among groups: the content of MMP-13 was significantly lower in group A than that in group B (P 0.01), and the content of MMP-1 + MMP-3 in group B was significantly lower than that in group B (P 0.05).Conclusion: the treatment of knee osteoarthritis with knee arthralgia may delay the destruction of cartilage and synovium by inhibiting the activity of MMP-1, MMP-3 and MMP-13, so as to slow down the damage to joint tissue and play a role in the treatment of knee osteoarthritis.
