【摘要】：目的:觀察電針"印堂""天樞"穴對腸易激綜合征(IBS)大鼠行為學及結腸辣椒素受體(TRPV 1)表達的影響,探討電針不同主治功能的穴位對身心疾病不同癥狀療效的差異及其相關機制。方法:Wistar新生大鼠32只隨機分為空白對照組、模型對照組、印堂組、天樞組。除空白對照組外,均采用新生期母子分離加醋酸灌腸并聯合結直腸擴張刺激法制備IBS大鼠模型。大鼠9周齡時印堂和天樞組分別給予電針干預,每次20min,隔日1次,共5次。采用腹部回撤反射對大鼠進行內臟敏感性評估,觀察第1次收縮波潛伏期及90s內收縮波個數;采用曠場實驗評估大鼠情緒心理行為,觀察其水平及垂直運動次數;采用免疫組織化學法檢測結腸組織TRPV 1的表達。結果:與空白對照組比較,模型對照組第1次收縮波潛伏期明顯縮短(P0.01),收縮波個數明顯增多(P0.01),水平運動次數以及垂直運動次數均明顯減少(P0.01),結腸內TRPV 1免疫反應物陽性表達水平增強(P0.05)。與模型對照組比較,天樞組及印堂組第1次收縮波潛伏期延長及90s內收縮波個數明顯減少(P0.01,P0.05),天樞組垂直運動次數增多(P0.05),印堂組的垂直運動次數及水平運動次數均明顯增多(P0.01),天樞組及印堂組結腸TRPV 1免疫反應物陽性表達顯著下降(P0.01,P0.05)。與印堂組比較,天樞組TRPV 1免疫反應物陽性表達顯著下降(P0.01)。結論:電針"印堂""天樞"穴均可明顯緩解IBS大鼠的腹痛并改善精神情緒的異常,電針"印堂"穴能更好地緩解模型大鼠抑郁樣情緒行為,而電針"天樞"穴治療腹痛效果較佳,二者對結腸內TRPV 1免疫反應物陽性表達的影響存在明顯差異。
[Abstract]:Objective: to observe the effect of electroacupuncture on behavior and expression of colonic capsaicin receptor TRPV1 in rats with irritable bowel syndrome (IBS).To explore the effect of electroacupuncture on different symptoms of physical and mental diseases and its related mechanism.Methods Thirty-two newborn Wistar rats were randomly divided into three groups: blank control group, model control group, Yantang group and Tianshu group.In addition to the blank control group, IBS rat model was established by the method of neonatal mother and child separation and acetic acid enema combined with colorectal dilatation stimulation.The rats were treated with electroacupuncture for 20 min, once every other day, for 5 times, at the age of 9 weeks in Yantang group and Tianshu group.The visceral sensitivity of rats was evaluated by abdominal withdrawal reflex, the latency of the first systolic wave and the number of systolic waves in 90s were observed, the emotional psychological behavior of rats was evaluated by open field experiment, and the horizontal and vertical movement times were observed.Immunohistochemical method was used to detect the expression of TRPV 1 in colon tissue.Results: compared with the blank control group,In the model control group, the latency of the first contraction wave shortened significantly, the number of the contraction wave increased significantly, the number of horizontal and vertical movements decreased significantly, and the positive expression of TRPV 1 immunoreactive substance increased in the colon.Compared with the model control group,In Tianshu group and Yantang group, the latency of the first systolic wave prolonged and the number of contraction waves decreased significantly in 90 s. The number of vertical motion increased in Tianshu group, and the number of vertical motion and horizontal motion increased significantly in Tianshu group, and the number of vertical motion and horizontal motion in Yantang group increased significantly (P 0.01), and in Tianshu group and in Tianshu group, the number of vertical motion and horizontal motion increased significantly; andThe positive expression of TRPV 1 immunoreactive substance in the colon of Yintang group decreased significantly.Compared with Yintang group, the positive expression of TRPV 1 immunoreactive substance in Tianshu group decreased significantly (P 0.01).Conclusion: electroacupuncture at "Yantang" and "Tianshu" can obviously relieve abdominal pain and improve mental mood abnormality in IBS rats. Electroacupuncture at "Yantang" can better relieve depressive emotional behavior in model rats.The effect of electroacupuncture on abdominal pain was better. There was significant difference between them on the expression of TRPV 1 immunoreactive substance in colon.
