穴位埋線對(duì)血管性癡呆模型大鼠學(xué)習(xí)記憶及海馬SOD、MDA的影響

【摘要】:目的:觀察百會(huì)、腎俞穴位埋線對(duì)血管性癡呆模型大鼠學(xué)習(xí)記憶、神經(jīng)細(xì)胞及海馬SOD、MDA的影響,探索百會(huì)、腎俞穴位埋線是否有腦神經(jīng)保護(hù)作用及抗氧化作用,為血管性癡呆治療提供新的依據(jù)。方法:造模前行水迷宮實(shí)驗(yàn)排除特異性大鼠,隨機(jī)假手術(shù)組10只,以經(jīng)改良的 Pulsinelli' s4 血管阻斷法(four-vesselocclusion,4-VO)建立 VD 大鼠模型[1]后隨機(jī)數(shù)表法分為VD模型組、穴位埋線組、針刺組。埋線組取穴:腎俞(雙)、百會(huì),每隔7天埋線1次,共埋2次;針刺組:治療之日起每日上午8時(shí)到12時(shí)之間進(jìn)行針刺,百會(huì),神庭穴向后斜刺,大椎,足三里,直刺平補(bǔ)平瀉手法捻轉(zhuǎn)1分鐘,后留針20分鐘,針刺每日一次,共針刺16天。模型組:造模成功后除常規(guī)飼養(yǎng)每日消毒觀察大鼠日常行為外不予任何治療處理。假手術(shù)組:麻醉后,行背側(cè)頸正中切口,不進(jìn)行椎動(dòng)脈燒灼,再行腹側(cè)頸正中切口,分離雙側(cè)頸總動(dòng)脈,穿線備用,但不進(jìn)行頸總動(dòng)脈夾閉,24小時(shí)候縫合切口。治療期間常規(guī)飼養(yǎng)每日消毒觀察大鼠日常行為外不予任何治療處理。以上動(dòng)物均在造模后、治療結(jié)束后行Morris水迷宮檢測(cè)各組大鼠學(xué)習(xí)記憶能力,水迷宮試驗(yàn)后隨機(jī)抽取各組中大鼠處死取含海馬的腦組織檢測(cè)細(xì)胞凋亡情況及SOD,MDA。結(jié)果;經(jīng)水迷宮實(shí)驗(yàn)檢測(cè)VD模型組表現(xiàn)出明顯的學(xué)習(xí)記憶障礙,SOD活性下降,MDA含量上升,與假手術(shù)組相比差異有統(tǒng)計(jì)學(xué)意義(P0.01);埋線組與針刺組兩組與模型組相比治療后SOD活性均有所上升,MDA含量均有所下降,且尋找跳臺(tái)次數(shù)明顯增多,潛伏期縮短,但埋線組優(yōu)于針刺組。各組抽取的大鼠腦切片做HE染色,光學(xué)顯微鏡下辨識(shí)海馬錐體細(xì)胞變化。發(fā)現(xiàn)假手術(shù)組錐體細(xì)胞排行緊湊,整體構(gòu)架規(guī)整,核清晰可見并深染。模型組則呈彌散空泡樣變大,核仁縮小。埋線組和針刺組細(xì)胞空泡彌散較模型組少,細(xì)胞構(gòu)架較清楚,核仁也可見。結(jié)論:百會(huì)、腎俞穴位埋線可以改善大鼠的學(xué)習(xí)記憶能力,提高大鼠大腦SOD活性,降低MDA含量,對(duì)大鼠腦組織損傷具有明顯的保護(hù)作用。
[Abstract]:Objective: to observe the effects of catgut embedding at Baihui and Shenshu points on learning and memory, nerve cells and MDA in hippocampus of rats with vascular dementia. Whether the Shenshu acupoint catgut embedding has the brain nerve protection and the antioxidation function, provides the new basis for the vascular dementia treatment. Methods: the water maze experiment before making the model excludes the specific rats, 10 rats in the random sham-operation group. Establishment of VD rat model by modified four-vessel inclusion 4-VO4 vessel occlusion method. [(1) the later random number table method was divided into VD model group, acupoint embedding group and acupuncture group. The acupoints in the catgut embedding group were collected: Shenshu (Shuangli, Baihui, once every 7 days), buried twice; Acupuncture group: daily acupuncture between 8:00 and 12:00 every day from the day of treatment, Baihui, Shenting point backward oblique prick, Dazhui, Zusanli, straight prick Pingbu Pingxiao technique twirled for 1 minute, then kept needle for 20 minutes. Acupuncture once a day for 16 days. Model group: after the model was successfully made, there was no treatment except routine feeding and daily disinfection to observe the daily behavior of the rats. The sham operation group: after anesthesia, the dorsal cervical median incision was performed. No cauterization of vertebral artery, ventral median cervical incision, separation of bilateral common carotid artery, puncture line reserve, but no common carotid artery occlusion. 24 hours suture incision. During the treatment period, routine feeding, disinfection and observation of the daily behavior of the rats were not treated with any treatment. All the above animals were treated after the establishment of the model. After the treatment, the learning and memory ability of the rats in each group was measured by Morris water maze. After the water maze test, the rats in each group were randomly selected and killed to detect apoptosis and SOD in the hippocampus. MDA. results; In VD model group, the activity of SOD decreased and the content of MDA increased by water labyrinth test. The difference was statistically significant compared with that of sham operation group (P 0.01). Compared with the model group, the activity of SOD in the catgut embedding group and acupuncture group decreased, and the frequency of searching for the platform increased and the incubation period was shortened. But the embedding group was superior to the acupuncture group. The brain slices of each group were stained by HE and the changes of hippocampal pyramidal cells were identified under optical microscope. The results showed that the pyramidal cells in the sham operation group were compact and the whole structure was regular. The nuclei were clearly visible and deeply stained. The model group showed diffuse vacuoles and reduced nucleoli. The number of vacuoles in the embedding group and acupuncture group was less than that in the model group. The cell structure was clear and the nucleolus was also visible in the model group. Conclusion: Baihui can be seen. The catgut embedding at Shenshu acupoint can improve the learning and memory ability of rats, increase the activity of SOD and decrease the content of MDA in the brain of rats. It has obvious protective effect on brain tissue injury in rats.
