【摘要】：目的:通過動(dòng)物實(shí)驗(yàn)研究,觀察參地補(bǔ)腎膠囊對(duì)單側(cè)輸尿管梗阻(unilateal ureteral obstruction,UUO)所致腎間質(zhì)纖維化大鼠模型腎功及Snail、E-cadherin蛋白表達(dá)的影響,探討參地補(bǔ)腎膠囊保護(hù)腎功能、抗腎間質(zhì)纖維化的作用機(jī)理。方法:將70只雄性SD大鼠隨機(jī)分為空白組、假手術(shù)組、模型組、尿毒清組、參地補(bǔ)腎組,每組14只。采用單側(cè)輸尿管結(jié)扎的方法建立腎間質(zhì)纖維化模型,證實(shí)造模成功后開始灌胃,尿毒清組給予尿毒清顆粒混懸液灌胃,參地補(bǔ)腎組給予參地補(bǔ)腎膠囊混懸液灌胃,空白組、假手術(shù)組、模型組均用等量生理鹽水灌胃,連續(xù)灌胃28天。觀察治療前后大鼠的一般狀態(tài),Scr、BUN的變化;將腎組織病理切片行HE染色,觀察腎臟病理學(xué)改變;采用免疫組化法檢測(cè)腎組織中Snail、E-cadherin表達(dá)的情況。結(jié)果:參地補(bǔ)腎組大鼠的一般狀態(tài)明顯好于模型組;參地補(bǔ)腎組大鼠腎臟病理改變明顯輕于模型組;參地補(bǔ)腎組大鼠在第35天血清Scr、BUN均明顯降低,與模型組比較有統(tǒng)計(jì)學(xué)意義(P0.05);參地補(bǔ)腎組大鼠在第35天腎組織中Snail表達(dá)明顯降低,E-cadherin表達(dá)明顯升高,與模型組比較有統(tǒng)計(jì)學(xué)意義(P0.05)。結(jié)論:參地補(bǔ)腎膠囊在一定程度上改善UUO大鼠的一般狀態(tài),降低大鼠Scr、BUN水平;參地補(bǔ)腎膠囊能減輕UUO大鼠腎臟病理學(xué)改變;參地補(bǔ)腎膠囊可能通過下調(diào)Snail的表達(dá),上調(diào)E-cadherin的表達(dá),在一定程度上延緩腎間質(zhì)纖維化的進(jìn)展。
[Abstract]:Objective: through animal experiments, observe ginseng Bushen Capsule on unilateral ureteral obstruction (unilateal ureteral obstruction, UUO) induced renal interstitial fibrosis in rat model of renal function and the expression of Snail and E-cadherin protein of ginseng, kidney capsule protect renal function and antitumor mechanism of renal interstitial fibrosis. Methods: 70 male SD rats were randomly divided into the blank group, the sham operation group, the model group, the uremia group and the Shen Di kidney tonifying group, with 14 rats in each group. Methods unilateral ureteral ligation of renal interstitial fibrosis model, confirmed after the success of modeling to gavage, Niaoduqing group with Niaoduqing granule suspension gavage, ginseng ginseng Bushen group given Bushen capsule suspension gavage, blank group, sham operation group, model group with normal saline intragastric gavage for 28 days. The changes of general condition, Scr and BUN in rats before and after treatment were observed. The pathological changes of renal tissue were observed by HE staining, and the expression of Snail and E-cadherin in renal tissue was detected by immunohistochemistry. Results: the general state of ginseng kidney group rats was significantly better than the model group; ginseng kidney group rat kidney pathological changes were obviously less than that of the model group; ginseng kidney group rats after thirty-fifth days of serum Scr, BUN were significantly decreased, had statistical significance compared with the model group (P0.05); ginseng kidney group the rats in the thirty-fifth days of renal tissue Snail expression was significantly reduced, E-cadherin expression was significantly increased and was statistically significant compared with the model group (P0.05). Conclusion: Ginseng Bushen capsule improving the general condition of UUO rats to a certain extent, reduce the level of Scr and BUN rats; ginseng Bushen capsule can alleviate the pathological changes of renal pathology in UUO rats; ginseng Bushen capsule could downregulate the expression of Snail, increase the expression of E-cadherin and delay the progression of renal interstitial fibrosis in to a certain extent.
