【摘要】：目的:以急性胃黏膜損傷大鼠為研究對(duì)象,觀察艾灸足三里、中脘穴對(duì)急性胃黏膜損傷模型大鼠胃黏膜三葉因子(TFF1/2/3)及下頜下腺三葉因子(TFF2/3)蛋白表達(dá)的影響,探討艾灸促進(jìn)胃黏膜增值修復(fù)的可能機(jī)制,為艾灸治療胃黏膜損傷理論在臨床的應(yīng)用中提供部分實(shí)驗(yàn)依據(jù)。方法:40只SPF級(jí)SD大鼠隨機(jī)分為4組:A.正常組、B.模型組、C.艾灸穴位組、D.艾灸非穴位組,首先對(duì)實(shí)驗(yàn)大鼠進(jìn)行艾灸足三里、中脘及其對(duì)照點(diǎn)預(yù)處理8天,然后用無(wú)水乙醇灌胃的方法造成急性胃黏膜損傷大鼠模型,肉眼觀察大鼠胃黏膜損傷情況,Guth法計(jì)算大鼠胃黏膜損傷指數(shù),光鏡下觀察胃黏膜組織形態(tài)學(xué)變化,免疫組化法檢測(cè)胃黏膜中TFF 1/2/3的含量,Western Blot法檢測(cè)下頜下腺TTT2/3的蛋白表達(dá)。結(jié)果:1,與正常組大鼠相比,模型組大鼠胃黏膜表面可見(jiàn)明顯片狀潰瘍形成,潰瘍部位上附著有大小不等的血痂,成點(diǎn)狀、線狀或條帶狀出血點(diǎn);與模型組相比,艾灸穴位組大鼠胃黏膜表面色澤較為紅潤(rùn),未見(jiàn)明顯片狀潰病形成,可見(jiàn)少數(shù)點(diǎn)狀出血;艾灸非穴組大鼠胃黏膜組織表面可見(jiàn)明顯充血點(diǎn),部分胃黏膜表面有血癡附著,并有潰瘍點(diǎn)形成。2,與正常組大鼠相比,模型組大鼠胃黏膜損傷指數(shù)顯著升高(P0.01);與模型組大鼠相比,艾灸穴位組大鼠胃黏膜損傷指數(shù)顯著降低(P0.01),與正常組無(wú)顯著性差異;與艾灸穴位組相比,艾灸非穴組大鼠的胃黏膜損傷指數(shù)較高(P0.01),與模型組無(wú)顯著性差異,光鏡下可見(jiàn)組織形態(tài)學(xué)的病理改變。3,與正常組相比,模型組大鼠胃黏膜中TFF1、TFF2、TFF3的含量均明顯增高(P0.05或P0.01);與模型組比較,艾灸穴位組大鼠胃黏膜中TFF1、TF F3的含量降低(P0.05),TFF2的含量雖與模型組相比無(wú)顯著性差異(P0.05),但也有所降低,艾灸穴位組大鼠胃黏膜中TFF1、TFF2、TFF3與正常組基本一致,無(wú)顯著性差異(P0.05);與艾灸穴位組比較,艾灸非穴位組大鼠胃黏膜中TFF1、TFF2、TFF3的含量雖無(wú)顯著性差異(P0.05),但也有所升高,與模型組相比,艾灸非穴位組大鼠TFF1的含量顯著降低(P0.05),TFF2、TFF3的含量雖無(wú)顯著性差異(P0.05),但也有所輕微降低。4,與正常組相比,模型組大鼠下頜下腺中TFF2、TFF3的表達(dá)均增高(p0.05或P0.01);與模型組相比,艾灸穴位組大鼠下頜下腺中TFF2、TFF3的蛋白表達(dá)均明顯降低(p0.05),艾灸穴位組大鼠下頜下腺中TFF2、TFF3的蛋白表達(dá)與正常組相比無(wú)顯著性差異(P0.05);與艾灸穴位組相比,艾灸非穴位組大鼠下頜下腺中TFF2、TFF3的蛋白表達(dá)雖無(wú)顯著性差異(P0.05),但也有所升高,與模型組相比,艾灸非穴位組大鼠下頜下腺中TFF2、TFF3的蛋白表達(dá)均明顯降低(p0.05),艾灸非穴位組大鼠下頜下腺中TFF2、TFF3的蛋白表達(dá)與正常組相比無(wú)顯著性差異(P0.05)。結(jié)論:1,艾灸預(yù)處理對(duì)大鼠急性胃黏膜損傷具有一定的保護(hù)作用。2,艾灸預(yù)處理對(duì)大鼠急性胃黏膜損傷的保護(hù)作用與胃黏膜中TFF1/2/3的表達(dá)有關(guān),說(shuō)明TFF可誘導(dǎo)胃黏膜細(xì)胞的增值修復(fù)。3,艾灸預(yù)處理對(duì)大鼠急性胃黏膜損傷的保護(hù)作用與下頜下腺中TFF2/3的蛋白表達(dá)有關(guān)。
[Abstract]:Objective: to acute gastric mucosal injury in rats as the research object, the observation of moxibustion at Zusanli and Zhongwan on trefoil factor of gastric mucosa in rats with acute gastric mucosal injury model (TFF1/2/3) and submandibular gland trefoil factor (TFF2/3) protein expression, to study the effect of moxibustion on gastric mucosa and promote value-added mechanism may provide some experimental repair. Based on the damage theory of gastric mucosa in the clinical application of moxibustion therapy. Methods: 40 SPF SD rats were randomly divided into 4 groups: A. normal group, B. model group, C. group, D. moxibustion moxibustion acupoint and non acupoint group, firstly, moxibustion at Zusanli on rats and control Zhongwan point pretreatment method for 8 days, and then ethanol gavage caused by acute gastric mucous membrane injury rat model of gastric mucosa injury in rats were observed to calculate the index of gastric mucosal injury in rats by Guth, the morphological changes of gastric mucosa were observed under light microscope, immunohistochemistry was used to detect the content of 1/2/3 TFF in gastric mucosa, gland TTT2/3 protein expression by Blot under mandibular Western. Results: 1 compared with normal rats, gastric mucosal surface model rats showed patchy ulcer, ulcer attached with size ranging from blood scab, punctate, linear or banded bleeding; compared with the model group, the surface color of gastric mucosa in rats of moxibustion acupoint group is not rosy, obvious flake ulcer formation, showing a few petechial hemorrhage; moxibustion non gastric mucosa surface of rats in acupuncture group showed obvious congestion, part of the surface of gastric mucosa blood crazy attachment, and ulcer formation. 2, compared with normal rats, gastric mucosal injury index of rats in model group were significantly increased (P0.01); compared with the model group rats, gastric mucosal injury index of Acupoint Moxibustion group rats were significantly decreased (P0.01), and no significant difference compared with normal group; moxibustion group, gastric mucosal injury index was higher moxibustion non acupoint group rats (P0.01), no significant difference with the model group, the pathological changes of tissue under visible light. 3, compared with the normal group, the rats in model group, TFF2 and TFF1 in gastric mucosa of TFF3 were significantly increased (P0.05 or P0.01); compared with the model group, Acupoint Moxibustion group and reduce the content of gastric mucosa of rats in TFF1, TF F3 (P0.05), the content of TFF2 was no significant compared with the model group the difference (P0.05), but also decreased, Acupoint Moxibustion group in gastric mucosa of rats in TFF1, TFF2, TFF3 and normal group, no significant difference (P0.05); compared with non Acupoint Moxibustion group, moxibustion acupoint group no content in gastric mucosa of rats in TFF1, TFF2 and TFF3 were significantly the difference (P0.05), but also increased, compared with the model group, the content of moxibustion and non acupoint group rats significantly decreased TFF1 (P0.05), there was no significant difference in TFF2, TFF3 (P0.05), but also has a slight decrease. 4, compared with the normal group, the expression of TFF2 and TFF3 of the mandibular glands of rats in model group were increased (P0.05 or P0.01); compared with the model group, TFF2 gland and TFF3 protein expression were significantly lower in Acupoint Moxibustion group rat submandibular (P0.05), Acupoint Moxibustion group rat submandibular glands TFF2, TFF3 protein expression compared with normal group no significant difference (P0.05); compared with moxibustion moxibustion group, non acupoint group expression in submandibular glands of rats TFF2, TFF3 protein have no significant difference (P0.05), but also increased, compared with the model group, TFF2 and TFF3 in gland protein expression decreased significantly in rat submandibular Ai Jiufei acupoint group (P0.05), Ai Jiufei acupoint group rat submandibular gland TFF2, TFF3 protein expression compared with normal group no significant difference (P0.05). Conclusion: 1, moxibustion preconditioning has a certain protective effect on acute gastric mucosal injury in rats. 2, the protective effect of moxibustion pretreatment on acute gastric mucosal injury in rats is related to the expression of TFF1/2/3 in gastric mucosa, indicating that TFF can induce gastric epithelial cell proliferation repair. 3, the protective effect of moxibustion preconditioning on acute gastric mucosal injury in rats is related to the protein expression of TFF2/3 in the submandibular gland.
