WNT/β-catenin信號通路在不同癌癥發(fā)生中的作用研究

研究背景WNT/β-catenin信號通路在人類器官和腫瘤發(fā)生中發(fā)揮關(guān)鍵效應(yīng)。WNTs是一組能夠編碼分泌型富含半胱氨酸的糖蛋白,其作為信號分子用于調(diào)節(jié)不同階段的胚胎發(fā)育并參與維持組織穩(wěn)態(tài)。WNT信號的畸變或異常將導(dǎo)致多種人類疾病,如白血病,先天性四肢切斷癥,精神分裂癥,腎損傷,骨發(fā)病率,肺纖維化和不同種類的癌癥。依據(jù)是否涉及β-catenin,WNT信號通路可分為典型(WNT/β-catenin途徑)和非典型途徑(WNT/平面細(xì)胞極性(PCP)或WNT/Ca2+途徑)。目前,最廣泛研究的WNT途徑是典型WNT/β-catenin途徑,其在癌癥起始和進(jìn)展中發(fā)揮的作用。在WNT/β-catenin途徑中,WNT配體可以結(jié)合卷曲蛋白(frizzled,Frz)受體激活蓬亂蛋白(DSH),β-catenin磷酸化被抑制,導(dǎo)致細(xì)胞內(nèi)β-catenin積累和核易位,隨后β-catenin作為轉(zhuǎn)錄共激活因子與T細(xì)胞因子(TCF)/淋巴增強因子(LEF1)轉(zhuǎn)錄因子復(fù)合,并激活下游靶基因使之過表達(dá),如CyclinD1,c-MYC,PLA2G2A等。TCF/LEF家族是一組通過高遷移率基團結(jié)合DNA的轉(zhuǎn)錄...
