Notchl與Wnt/β-catenin信號(hào)通路調(diào)控肝癌干細(xì)胞“干性”的機(jī)制研究

目的:肝細(xì)胞肝癌(Hepatocellular Carcinoma, HCC)仍是嚴(yán)重威脅我國人民生命健康的常見惡性腫瘤,目前總體療效與患者生存率還不能令人滿意。其高度的異質(zhì)性是其較其他實(shí)體腫瘤更易發(fā)生轉(zhuǎn)移復(fù)發(fā)的主要原因。腫瘤的異質(zhì)性意味著腫瘤中存在一小部分具有“干性”潛能的腫瘤干細(xì)胞(Liver cancer stem cell, LCSCs),其具有許多如無限增殖、自我更新、免疫缺陷小鼠體內(nèi)成瘤以及自我分化和致瘤能力等干細(xì)胞生物學(xué)特性。現(xiàn)有研究發(fā)現(xiàn)CD133、CD9、CD13、CD24、CD44和上皮細(xì)胞粘附分子(EpCAM)等可作為肝癌干細(xì)胞(LCSCs)的表面標(biāo)記,而Notch、Wnt/β-catenin1-3 Hedgehog信號(hào)通路等常在干細(xì)胞的自我更新與分化發(fā)育中起重要作用。因此,本研究擬首先觀察肝癌標(biāo)本中上述不同LCSC表面標(biāo)記的表達(dá)與臨床預(yù)后及病理特征之間的關(guān)系。再檢測(cè)浮球培養(yǎng)富集的肝癌干細(xì)胞的表面標(biāo)記表達(dá)情況,并通過一系列“干性”鑒定實(shí)驗(yàn)和干性基因的檢測(cè)鑒定其“干性"特征,進(jìn)一步通過一組干預(yù)與調(diào)控研究探討Notch和Wnt/β-catenin信號(hào)通路在肝癌干細(xì)胞(L...
