miR-197與IL6/STAT3信號(hào)通路相互調(diào)控在肝癌生長(zhǎng)的作用及機(jī)制研究

慢性炎癥,無(wú)論是感染如肝炎、宮頸炎還是自身免疫疾病性質(zhì)的如結(jié)腸炎等,都與癌癥的發(fā)生密切相關(guān),慢性炎癥與腫瘤相互關(guān)系一直是生物醫(yī)學(xué)領(lǐng)域研究的熱點(diǎn)之一。STAT3(Signal transducer and activator of transcription3)作為介導(dǎo)炎癥刺激的重要效應(yīng)分子,在多種人類腫瘤包括人肝癌中被檢測(cè)到處于持續(xù)性活化狀態(tài),且與腫瘤的惡性侵襲及預(yù)后差呈正相關(guān)。然而,肝癌中除了活化的STAT3即磷酸化STAT3(p-STAT3)升高外,也有報(bào)導(dǎo)STAT3總蛋白水平有顯著上調(diào),但其上調(diào)機(jī)制卻不甚明了。本研究以人肝癌細(xì)胞為模型,體外模擬了炎性因子IL-6(Interleukin6)刺激后,人肝癌細(xì)胞內(nèi)發(fā)生的變化。結(jié)果顯示,IL-6刺激4小時(shí)內(nèi),STAT3總蛋白水平在其mRNA相對(duì)含量沒(méi)有顯著差異的情況下發(fā)生了明顯上調(diào),提示此過(guò)程中可能有microRNA (miRNA)參與,因?yàn)閙iRNA對(duì)蛋白水平的調(diào)節(jié)發(fā)生在轉(zhuǎn)錄后(post-transcriptional)水平。Microarray分析IL-6刺激前后肝癌細(xì)胞系HepG2中差異表達(dá)最顯著的前10種microRNAs,將其...
