離心力對(duì)質(zhì)譜流式細(xì)胞技術(shù)檢測(cè)結(jié)腸癌細(xì)胞表面抗原CD45表達(dá)結(jié)果的影響

【摘要】:目的探討離心力對(duì)質(zhì)譜流式細(xì)胞技術(shù)檢測(cè)結(jié)腸癌細(xì)胞表面抗原CD45表達(dá)結(jié)果的影響。方法將體外培養(yǎng)的結(jié)腸癌HCT116細(xì)胞分為400 g離心組和140 g離心組(每組細(xì)胞數(shù)均為3×10~6個(gè)),分別采用400 g、140 g離心力離心5 min,重復(fù)兩次,收集細(xì)胞。采用質(zhì)譜流式細(xì)胞技術(shù)檢測(cè)兩組死亡細(xì)胞百分比、抗人CD45金屬聚合抗體(154Sm)信號(hào)強(qiáng)度10~2的細(xì)胞數(shù)占總活細(xì)胞數(shù)的比例、t-SNE圖顯示活細(xì)胞中高表達(dá)CD45的細(xì)胞數(shù)量。結(jié)果 400 g離心力組及140 g離心力組死亡細(xì)胞百分比分別為(7.69±0.7)%、(19.06±0.66)%,兩組比較P0.01。400 g離心力組及140 g離心力組154Sm信號(hào)強(qiáng)度10~2的細(xì)胞數(shù)分別占活細(xì)胞總數(shù)的0.2%[(0.19±0.08)%]、2.9%[(2.88±0.03)%],兩組比較P0.01。t-SNE圖顯示,400 g離心力組及140 g離心力組CD45高表達(dá)細(xì)胞分別為(167.41±70.48)、(201.20±20.96)個(gè),兩組比較P0.01。結(jié)論應(yīng)用質(zhì)譜流式細(xì)胞技術(shù)檢測(cè)結(jié)腸癌細(xì)胞表面抗原CD45表達(dá)時(shí),140 g離心力分離的細(xì)胞檢測(cè)結(jié)果更好。
