【摘要】：目的探討血根堿對宮頸癌細(xì)胞增殖、凋亡、侵襲、遷移能力的影響機(jī)制。方法分別用MTT法、流式細(xì)胞儀、細(xì)胞劃痕實驗、Transwell小室檢測血根堿作用后的宮頸癌細(xì)胞增殖、凋亡、遷移、侵襲能力。Western blot檢測經(jīng)血根堿作用后宮頸癌細(xì)胞中E-Cadherin、PTEN、β-catenin、MMP2的蛋白表達(dá)水平。結(jié)果 0.6、0.8μmol/L血根堿對宮頸癌細(xì)胞增殖具有抑制作用。0.8μmol/L血根堿作用48h后宮頸癌細(xì)胞HeLa和Siha凋亡率高達(dá)(45.68±2.26)%和(31.89±3.80)%。0.8μmol/L血根堿作用3h后宮頸癌細(xì)胞HeLa和Siha黏附率僅有(67.45±2.13)%和(73.59±2.61)%。0.8μmol/L血根堿作用16h后宮頸癌細(xì)胞HeLa和Siha侵襲數(shù)目僅有(39.64±1.98)個和(43.87±2.83)個。經(jīng)血根堿作用后的宮頸癌細(xì)胞中E-Cadherin和PTEN的表達(dá)量增加,β-catenin和MMP2的表達(dá)量減弱。結(jié)論血根堿對宮頸癌細(xì)胞具有抑制增殖及促凋亡作用,其機(jī)制可能與黏附蛋白E-Cadherin、β-catenin和PTEN、MMP2有關(guān)。
