FK506結(jié)合蛋白5在肝細胞癌中的表達及其臨床意義

【摘要】:目的:探討FK506結(jié)合蛋白5(FKBP5)在肝細胞癌(HCC)中的表達及臨床意義。方法:用q RT-PCR和Western blot法檢測30對新鮮HCC及相應(yīng)癌旁組織中FKBP5 m RNA和蛋白的表達;免疫組化法檢測107例HCC患者石蠟標本中FKBP5的表達,并分析其與HCC臨床病理特征及預(yù)后的關(guān)系。結(jié)果:30例新鮮HCC標本中,26例(86.7%)HCC組織FKBP5 m RNA的表達水平明顯高于相應(yīng)癌旁組織,HCC組織中FKBP5蛋白表達量明顯高于其癌旁組織。石蠟標本分析顯示,FKBP5蛋白在HCC組織中的高表達率明顯高于癌旁組織(63.6%vs.28.0%,P0.001);FKBP5的表達水平與腫瘤大小(P=0.009)、腫瘤個數(shù)(P=0.018)、微血管侵犯(P=0.020)、TNM分期(P=0.009)和BCLC分期(P=0.024)有關(guān),且FKBP5高表達患者術(shù)后總體生存率和無瘤生存率均明顯低于低表達組(均P0.001);FKBP5高表達是HCC患者術(shù)后總體生存(HR=1.992,95%CI=1.127~3.522,P=0.018)和無瘤生存(HR=1.787,95%CI=1.067~2.992,P=0.027)的獨立危險因素。結(jié)論:FKBP5在HCC組織中高表達,且與患者不良臨床病理特征及預(yù)后密切相關(guān),提示其是一個潛在的HCC預(yù)后分子標志物。
[Abstract]:Objective: to investigate the expression and clinical significance of FK506 binding protein 5 (FKBP5) in hepatocellular carcinoma (HCC). Methods: the expression of FKBP5 m RNA and protein in 30 pairs of fresh HCC and corresponding paracancerous tissues were detected by Q RT-PCR and Western blot, and the expression of FKBP5 in paraffin specimens of HCC patients was detected by immunohistochemistry, and the relationship between FKBP5 and clinicopathological features and prognosis of HCC was analyzed. Results: the expression level of FKBP5 m RNA in 26 (86.7%) HCC tissues was significantly higher than that in corresponding paracancerous tissues, and the expression of FKBP5 protein in HCC tissues was significantly higher than that in paracancerous tissues.
