基于納米金的多功能納米系統(tǒng)及其腫瘤診療一體化應(yīng)用

【摘要】:納米金顆粒具有基于X射線的CT成像及放療增敏特性,但由于缺乏有效可靠的評估手段,因而限制了納米金的臨床應(yīng)用及X射線診療一體化。而如何通過簡單的構(gòu)筑方法高效整合多種腫瘤診斷和治療模式,依然是發(fā)展納米診療一體化制劑所面臨的機(jī)遇與挑戰(zhàn)。本文的目的是基于不同粒徑的球形納米金顆粒,研究X射線特性的尺寸效應(yīng),并以優(yōu)選粒徑的納米金顆粒為基礎(chǔ),結(jié)合其他功能顆粒構(gòu)建多功能納米系統(tǒng)應(yīng)用于腫瘤的診療一體化。具體研究內(nèi)容如下:1.可控制備3-50 nm范圍內(nèi)七種粒徑的球形納米金顆粒。結(jié)果表明,種子生長制備方法重復(fù)性好,所制備粒徑之間存在明顯差異,具備較好的單分散性,表面PEG修飾的納米金顆粒擁有較好的溫度穩(wěn)定性、p H穩(wěn)定性和膠體穩(wěn)定性,并具備較低的細(xì)胞毒性;2.納米金X射線特性的尺寸效應(yīng)研究。結(jié)果表明,不同粒徑納米金顆粒CT增強(qiáng)對比、吸收劑量跳數(shù)、相對電子密度及細(xì)胞水平的放療增敏效果不同,且隨粒徑的變化存在相似規(guī)律,13.2 nm納米金顆粒可同時具備最佳的CT增強(qiáng)和放療增敏特性;3.基于蒙卡模擬的尺寸效應(yīng)評估和理論研究。結(jié)果表明,蒙卡模擬仿真模型可充分還原納米金顆粒懸浮液混合物的特點(diǎn),納米金顆粒在Ke V和Me V射線能量下光子通量和吸收劑量隨粒徑的變化存在相似特點(diǎn),10-14 nm納米金顆粒可同時具備較好的射線衰減和劑量吸收,并提出相應(yīng)的假說及解釋;4.優(yōu)選納米金顆粒的X射線診療一體化研究。結(jié)果表明,13.2 nm優(yōu)選納米金顆粒具有生物安全性好、被動腫瘤靶向能力強(qiáng)、體內(nèi)循環(huán)時間長等優(yōu)點(diǎn),可同時實(shí)現(xiàn)實(shí)現(xiàn)CT增強(qiáng)成像和放射增敏治療,且效果優(yōu)于臨床成像劑碘海醇和臨床放療增敏劑甘氨雙唑鈉;5.基于優(yōu)選納米金顆粒的多功能診療一體化研究。結(jié)果表明,將13.2 nm優(yōu)選納米金顆粒與普魯士藍(lán)納米顆粒相結(jié)合,構(gòu)建一種具備核/衛(wèi)星結(jié)構(gòu)的多功能納米系統(tǒng)(CSNPs),其粒徑約為138 nm,實(shí)現(xiàn)T1/T2-MRI和CT的多模態(tài)成像,并實(shí)現(xiàn)光熱治療與放療增敏的協(xié)同治療,達(dá)到幾乎完全抑制腫瘤生長的目的;綜上所述,本文以納米金為基礎(chǔ),成功研究其作為X射線診療一體化的各種特性,并以此構(gòu)建多功能納米顆粒,有望作為一種有效的多功能納米制劑應(yīng)用于腫瘤的診療一體化領(lǐng)域。
