羅勒多糖抑制TGF-β誘導的A549細胞侵襲運動能力研究

【摘要】:目的羅勒多糖具有抗腫瘤侵襲轉移、化療增效以及降血糖、降血脂和抗血栓等多種生物活性作用。本研究探討羅勒多糖對轉化生長因子-β(transforming growth factor-beta,TGF-β)誘導的人肺癌A549細胞侵襲運動能力的抑制作用,揭示羅勒多糖抑制上皮間質轉化作用的可能機制。方法體外培養(yǎng)肺癌A549細胞,實驗分為3組:(1)正常對照組,不加任何處理;(2)模型組,TGF-β5ng/mL;(3)羅勒多糖組,TGF-β5ng/mL+羅勒多糖200μg/mL。免疫熒光檢測羅勒多糖對E-cadherin、Vimentin和α-SMA表達的影響,細胞侵襲實驗和運動實驗檢測細胞對人工基底膜的降解和運動能力,細胞劃痕實驗檢測細胞運動能力。結果與正常對照組比較,模型組細胞Vimentin和α-SMA表達增強,E-cadherin表達減弱。羅勒多糖處理后Vimentin和α-SMA表達減弱,E-cadherin表達增強。TGF-β可提高細胞降解人工基底膜的能力,單位視野下侵襲至下室的細胞數(shù)模型組為69.0±4.3,與正常對照組(42.0±4.30)和羅勒多糖組(37.0±4.41)比較,差異有統(tǒng)計學意義,均P0.05。TGF-β增強了細胞的侵襲能力,羅勒多糖可以抑制TGF-β誘導的A549細胞侵襲能力。TGF-β使FN趨化的遷移細胞數(shù)目增多,單位視野下侵襲至下室的細胞數(shù)模型組為95.2±3.83,與正常對照組(69.2±5.11)和羅勒多糖組(68.2±6.18)比較,差異均有統(tǒng)計學意義,均P0.05。劃痕實驗顯示,TGF-β增強了細胞的遷移能力,羅勒多糖可以抑制TGF-β誘導的A549細胞遷移能力。結論羅勒多糖能夠調(diào)節(jié)A549細胞上皮間質轉化,這可能是其抑制A549細胞的侵襲轉移能力的重要機制。
