miR-105-1在原發(fā)性肝細(xì)胞癌中表達(dá)的研究

【摘要】:目的定量分析miRNA-105-1(miR-105-1)在原發(fā)性肝細(xì)胞癌(HCC)及癌旁正常組織中表達(dá)水平的差異。方法取患者手術(shù)新鮮組織標(biāo)本(10對T-N配對組織和另外40例原發(fā)性肝癌組織)提取RNA,采用實時熒光定量PCR(RT-q PCR)技術(shù)檢測標(biāo)本中miR-105-1基因表達(dá)量。并下載公共基因表達(dá)數(shù)據(jù)庫(GEO)芯片數(shù)據(jù)加以驗證,通過2-ΔΔCt方法計算得到。結(jié)果實驗結(jié)果表明在10對配對的HCC組織中的miR-105-1的表達(dá)量顯著低于癌旁正常組織,差異有統(tǒng)計學(xué)意義(P=0.046)。在50個HCC組織的表達(dá)量顯著低于10個癌旁組織,差異有統(tǒng)計學(xué)意義(P=0.031)。GEO數(shù)據(jù)庫芯片GSE22058-GPL10457中包含96個配對的HCC與癌旁正常組織,miR-105-1在HCC中下調(diào),差異有統(tǒng)計學(xué)意義(P=0.035)。GSE21362中包含73個配對的HCC與癌旁正常組織,miR-105-1在HCC中也是下調(diào)的,差異有統(tǒng)計學(xué)意義(P=0.042)。結(jié)論 miR-105-1在原發(fā)性肝癌患者中較正常人群表達(dá)量低,可能是HCC患者發(fā)生發(fā)展的重要分子之一,并可以作為判斷預(yù)后的一個指標(biāo)和治療的一個新靶點。
