【摘要】：目的定量分析miRNA-105-1(miR-105-1)在原發(fā)性肝細胞癌(HCC)及癌旁正常組織中表達水平的差異。方法取患者手術(shù)新鮮組織標本(10對T-N配對組織和另外40例原發(fā)性肝癌組織)提取RNA,采用實時熒光定量PCR(RT-q PCR)技術(shù)檢測標本中miR-105-1基因表達量。并下載公共基因表達數(shù)據(jù)庫(GEO)芯片數(shù)據(jù)加以驗證,通過2-ΔΔCt方法計算得到。結(jié)果實驗結(jié)果表明在10對配對的HCC組織中的miR-105-1的表達量顯著低于癌旁正常組織,差異有統(tǒng)計學意義(P=0.046)。在50個HCC組織的表達量顯著低于10個癌旁組織,差異有統(tǒng)計學意義(P=0.031)。GEO數(shù)據(jù)庫芯片GSE22058-GPL10457中包含96個配對的HCC與癌旁正常組織,miR-105-1在HCC中下調(diào),差異有統(tǒng)計學意義(P=0.035)。GSE21362中包含73個配對的HCC與癌旁正常組織,miR-105-1在HCC中也是下調(diào)的,差異有統(tǒng)計學意義(P=0.042)。結(jié)論 miR-105-1在原發(fā)性肝癌患者中較正常人群表達量低,可能是HCC患者發(fā)生發(fā)展的重要分子之一,并可以作為判斷預后的一個指標和治療的一個新靶點。
[Abstract]:Objective to quantitatively analyze the expression of miRNA-105-1 (miR-105-1) in primary hepatocellular carcinoma (HCC) and adjacent normal tissues. Methods RNA, was extracted from fresh tissues of patients (10 pairs of T / N matched tissues and 40 cases of primary liver cancer).
