細胞自噬在姜黃素增強LoVo細胞對伊立替康化療敏感性中的作用

【摘要】:目的:探討姜黃素(Cur)增強結(jié)直腸癌LoVo細胞對伊立替康(CPT-11)化療敏感性的作用與細胞自噬的關(guān)系。方法:選擇人結(jié)腸癌LoVo細胞按照Cur及CPT-11的不同給藥方式,分為對照組(Con)、Cur單藥組(Cur)、CPT-11單藥組(Cpt)及混合給藥組(Mix)。Western blot法檢測自噬相關(guān)蛋白LC3-Ⅱ、P62的表達,細胞免疫熒光法檢測LC3-Ⅱ蛋白的表達;透射電鏡觀察自噬體的形成;自噬抑制劑3-甲基腺嘌呤(3-MA)預處理LoVo細胞之后,CCK-8法檢測不同給藥方式對細胞24 h生長抑制率的影響,檢測LC3-Ⅱ、P62蛋白表達的變化。結(jié)果:與Cur、Cpt組相比,Mix組中LC3-Ⅱ蛋白的表達顯著上調(diào),P62蛋白的表達顯著下調(diào);與Cur、Cpt組相比,Mix組中自噬體明顯增多;3-MA預處理后,Cur及CPT-11不同給藥方式對LoVo細胞的抑制率顯著降低,Mix組LC3-Ⅱ蛋白的表達顯著下調(diào),P62蛋白的表達顯著上調(diào)。結(jié)論:姜黃素可增強LoVo細胞對CPT-11的化療敏感性,其機制可能與激活細胞自噬有關(guān)。
