【摘要】：目的:研究鋅指轉(zhuǎn)錄因子580(ZNF580)在胃癌中的表達(dá)狀況,初步探討其在胃癌發(fā)生、發(fā)展過(guò)程中所扮演的的角色,包括其對(duì)于胃癌所造成的影響和血管內(nèi)皮生長(zhǎng)因子(VEGF)之間,潛存的相互聯(lián)系。方法:將山西醫(yī)科大學(xué)第一醫(yī)院2010年1月到2013年2月期間,通過(guò)手術(shù)切除的、病理確診的144例胃癌以及癌旁組織(距癌腫5cm)的石蠟樣本,作為試驗(yàn)研究對(duì)象,依據(jù)制作芯片的方法并整合免疫組織化學(xué)手段,測(cè)驗(yàn)ZNF580與VEGF蛋白在胃癌之中的蛋白表達(dá)程度。結(jié)果:ZNF580和VEGF在144例胃癌中的陽(yáng)性率(63.9%、57.6%)均明顯高于癌旁組(18.8%、13.9%)(P0.05)。在胃癌中ZNF580蛋白和VEGF蛋白的表達(dá)分別與腫瘤大小、浸潤(rùn)深度、TNM分期、淋巴結(jié)轉(zhuǎn)移有關(guān)(P0.05),而它們的表達(dá)與年齡、性別及分化程度無(wú)關(guān)(P0.05)。胃癌組織中ZNF580和VEGF表達(dá)正相關(guān)(P=0.005)。在單因素生存分析的結(jié)果中顯示,ZNF580和VEGF蛋白高表達(dá)與生存時(shí)間均表現(xiàn)為負(fù)相關(guān)(P0.05)。Cox多元回歸分析ZNF580陽(yáng)性胃癌的預(yù)后更差(RR=3.154,P0.05)和VEGF陽(yáng)性胃癌的預(yù)后更差(RR=4.094,P0.05)。結(jié)論:ZNF580蛋白在胃癌組織中表現(xiàn)為高表達(dá)的狀態(tài),且其在胃癌的發(fā)生和發(fā)展過(guò)程中可能與通過(guò)與VEGF起協(xié)同作用而使胃癌的預(yù)后更差,ZNF580結(jié)合VEGF也許可以用來(lái)做為預(yù)測(cè)胃癌預(yù)后的因子。
[Abstract]:Objective: to study the expression of zinc finger transcription factor 580 (ZNF580) in gastric cancer and its role in the development of gastric cancer, including its influence on gastric cancer and vascular endothelial growth factor (VEGF). Hidden interconnections. Methods: from January 2010 to February 2013, paraffin samples of 144 cases of gastric cancer and paracancerous tissues (5cm), which were surgically resected and pathologically confirmed, were taken as the subjects of the experiment, which were collected from the first Hospital of Shanxi Medical University from January 2010 to February 2013. The expression of ZNF580 and VEGF protein in gastric cancer was measured by the method of microarray and immunohistochemistry. Results: the positive rates of ZNF580 and VEGF in 144 cases of gastric cancer (63.9%, 57.6%) were significantly higher than those in the paracancerous group (18.8%, 13.9%) (P0.05). The expression of ZNF580 protein and VEGF protein were related to tumor size, depth of invasion, TNM stage and lymph node metastasis (P0.05), but their expression was not related to age, sex and differentiation degree (P0.05). There was a positive correlation between the expression of ZNF580 and VEGF in gastric cancer (P < 0. 005). The results of univariate survival analysis showed that there was a negative correlation between the high expression of ZNF580 and VEGF protein and survival time (P0.05). The prognosis of ZNF580 positive gastric cancer was worse than that of ZNF580 positive gastric cancer (RR=3.154,). The prognosis of VEGF-positive gastric cancer was worse than that of P05) (RR=4.094,P0.05). Conclusion: ZNF580 protein is highly expressed in gastric cancer tissues, and it may play a synergistic role with VEGF in the carcinogenesis and development of gastric cancer to make the prognosis of gastric cancer worse. ZNF580 combined with VEGF may be used as a predictor of prognosis in gastric cancer.
