應(yīng)用基于色譜質(zhì)譜聯(lián)用技術(shù)的蛋白質(zhì)組學(xué)鑒定篩選胃癌相關(guān)蛋白質(zhì)
[Abstract]:Objective to screen gastric cancer related proteins by proteomics based on LC-MS and to explore their functions. Methods from April 2011 to September 2012, 16 cases of gastric adenocarcinoma were diagnosed and surgically resected from the fourth Hospital of Hebei Medical University. The differentially expressed proteins in gastric cancer tissues and adjacent normal tissues were screened by proteomics identification based on LC-MS, and the differentially identified proteins were verified by immunohistochemical staining. Results the results of proteomics analysis based on LC-MS showed that 3 000 ~ 4 000 proteins were identified in gastric cancer tissues and adjacent normal tissues, among which 12 proteins had significant differences. The results were compared with GO database. Removing structural proteins classified as "cellular components" in secondary nodes, four proteins that might be associated with gastric cancer were found in the "molecular function" and "metabolic processes". They are glutathione mercaptotransferase-蟺 -1 (GST- 蟺 -1), copper zinc superoxide dismutase (Cu/Zn-SOD), pyruvate kinase M2 (PK-M2) and P120 catenin (P120-ctn). The results of immunohistochemical staining showed that the positive rate of GST- 蟺 -1 PK-M2P P120-ctn was higher than that of adjacent normal tissues, and the positive rate of Cu/Zn-SOD was lower than that of adjacent normal tissues (P0.001). The results were consistent with the results of proteomics identification based on LC-MS. Conclusion the proteome identification based on LC-MS can screen out GST- 蟺 -1CU / Zn-SODN-PK-M2P120-ctn, which is related to many cell biological activities of gastric cancer. It is confirmed that proteomics based on LC-MS is of great value in the screening of gastric cancer related proteins.
