OLC1蛋白在NSCLC患者組織中的表達情況及其與預后的關系

【摘要】:背景與目的OLC1(overexpressed in lung cancer 1)是一個新型肺癌相關基因,前期的研究發(fā)現(xiàn),在肺鱗癌、腺癌、小細胞癌組織中均有較高表達,OLC1蛋白在肺癌患者外周血漿中的表達顯著高于正常人,并可作為肺癌的輔助診斷的候選標志物。OLC1在食管鱗癌、卵巢癌、乳腺癌、結直腸癌組織中也有較高表達,并與不良預后相關,但OLC1與肺癌患者預后的關系一直缺乏相關研究。本研究通過檢測OLC1在非小細胞肺癌(NSCLC)患者癌組織中的蛋白表達情況,結合預后資料,分析OLC1的過表達與肺癌患者預后的關系。方法分別對包含75例鱗癌和90例腺癌的癌組織和癌旁組織的組織芯片以及33例鱗癌組織切片進行免疫組化染色,檢測OLC1蛋白的表達水平,分析其在組織中的表達與臨床特征及預后的關系。結果OLC1在腺癌患者癌組織中的過表達率(87.5%)明顯高于鱗癌(55.3%,P0.001)。OLC1在腺癌患者癌組織中的過表達與預后無顯著相關性,在鱗癌組織中的過表達與不良預后相關,單因素Kaplan-Meier生存分析提示OLC1過表達的患者具有更差的預后(P=0.042),多因素Cox回歸分析顯示兩組之間的生存差異無統(tǒng)計學意義(HR=2.048,95%CI 1.00-4.19,P=0.05)。OLC1在年齡≥60歲的患者中更易出現(xiàn)過表達(P=0.002),OLC1與年齡≥60歲共同作為危險因素時,患者的預后顯著變差(HR=5.125,95%CI 1.756-14.96,P=0.003)。結論OLC1在腺癌組織中的過表達率明顯高于鱗癌,但OLC1在癌組織中的過表達與腺癌患者的預后關系不明顯;OLC1在≥60歲的鱗癌患者組織中更易表達,并可能與年齡一起共同影響鱗癌患者的預后,但在癌組織中的過表達不足以作為影響肺鱗癌患者預后的獨立危險因素。

Previous studies have found that it is highly expressed in squamous cell carcinoma, adenocarcinoma and small cell carcinoma. The expression of OLC1 protein in peripheral plasma of lung cancer patients was significantly higher than that in normal controls, and could be used as a candidate marker for auxiliary diagnosis of lung cancer. OLC1 was also highly expressed in esophageal squamous cell carcinoma, ovarian cancer, breast cancer and colorectal cancer. But the relationship between OLC1 and prognosis of lung cancer patients has not been studied. In this study, we examined the expression of OLC1 protein in non-small cell lung cancer (NSCLC) patients with (NSCLC), and analyzed the relationship between the overexpression of OLC1 and the prognosis of NSCLC patients.
